By participation in the transport of nitrate of paddy rice and especially under the condition short of nitrogen and poor pHenvironment, the analysis of mRNA expression shows that OsNRT2.3a is induced by low nitrate nitrogen so that the expression only occurs in lateral roots; and OsNRT2.3b has expressions in root system and parts over the ground and is responsible for the transport of nitrate. The OsNRT2.3a and OsNRT2.3b proteins are expressed in a frogspawn heterogenous system and determined as high compatible nitrate transport protein. A pH regulation and control site is found to exist in both proteins which are regulated and controlled by pH. Under low nitrogen condition, OsNRT2.3 transgenic plant shows higherefficiency in transport of nitrate, thus improving the utilization efficiency of nitrogen and the ultimate output.

A preparation method of the stent comprises the following steps of: (1) processing the porous titanium or porous titanium oxide or porous titanium alloy to a shape and a size required by the subchondral bone layer; (2) preparing cartilage layer slurry; and (3) molding and treating a stent, putting the subchondral bone layer prepared in the step (1) into a mold, injecting the cartilage layer slurry prepared in the step (2) into the mold, putting the mold into a freeze dryer, freezing for at least 5 hours, molding, performing thermal crosslinking and chemical crosslinking on the molded stent, and putting the chemically-crosslinked stent into sterile water for rinsing to obtain the stent for osteochondral defect repair.

The preparation method comprises the following steps of: soaking a titanium sample subjected to micro-arc oxidation treatment in an alkaline solution, and carrying out hydrothermal treatment to form a hydroxyapatite bioactive coating on the surface of micro-arc titanium oxide; putting an aluminum template into a culture dish, then adding a mixture of a prepolymer of PDMS and a curing agent, and curing to obtain a PDMS template; and mixing a chitosan aqueous solution and a sodium alginate aqueous solution, adding a drug to obtain a drug-loaded hydrogel solution, adding a photoinitiator to the drug-loaded hydrogel solution, dropwise adding to the template, placing a sample on the PDMS template, curing under ultraviolet light, and carrying out freeze drying. According to the method, a composite coating with a porous structure and a nanorod shape is generated through micro-arc oxidation and hydrothermal treatment, and the bonding strength and the biological activity of the implant and bones are improved. The drug-loaded composite hydrogel is loaded on the surface of the micro-arc titanium oxide through the PDMS template, so that the implant coating is endowed with more biological functions.

MN Gene and Protein

Identified herein is the location of the MN protein binding site, and MN proteins / polypeptides that compete for attachment to vertebrate cells with immobilized MN protein. Such MN proteins / polypeptides prevent cell-cell adhesion and the formation of intercellular contacts. The MN protein binding site is a therapeutic target that can be blocked by organic or inorganic molecules, preferably organic molecules, more preferably proteins / polypeptides that specifically bind to that site. Therapeutic methods for inhibiting the growth of preneoplastic / neoplastic vertebrate cells that abnormally express MN protein are disclosed. Vectors are provided that encode the variable domains of MN-specific antibodies and a flexible linker polypeptide separating those domains. Further vectors are disclosed that encode a cytotoxic protein / polypeptide operatively linked to the MN gene promoter or a MN gene promoter fragment comprising the HIF-1 consensus binding sequence, and which vectors preferably further encode a cytokine. The MN gene promoter is characterized, and the binding site for a repressor of MN transcription is disclosed. Further, the hypoxia inducibility of the MN gene and the uses of such inducibility are disclosed.
