The invention discloses a separation and purification method of sweet corn cob
polysaccharide. The separation and purification method comprises the following steps of
drying and crushing sweet corn cobs, adding normal
hexane into sieved sweet corn cobs according to the
weight volume ratio being 1: (8 to 12), performing magnetic stirring under the
room temperature condition for 1 to 1.5 hours, and centrifuging at the speed of 4000r / min for 10 to 15 minutes; performing
hot water extraction and centrifuging on degreased sweet corn cobs under the conditions that the temperature is 100DEG C, the material-to-
liquid ratio is 1:(18 to 22), and the time is 2.5 to 3.5 hours, and taking supernate; adding 3 to 5g of
ammonium sulfate into a 25mL colorimetric tube according to an isopropanol-
ammonium sulfate aqueous two-
phase diagram, and then adding sweet corn cob crude
polysaccharide liquid; after the
ammonium sulfate is completely dissolved, finally adding the isopropanol until the volume is 20ml, so as to form an aqueous two-phase
system solution; enabling the aqueous two-phase
system solution processed by ultrasonic
waves to stand in a separating funnel for phase separation; respectively determining the volumes of upper and lower phases, and the contents of polysaccharides and
protein of the upper and lower phases; preserving the
polysaccharide solution obtained after separating and purifying two aqueous phases at 4DEG C in a refrigerator for 1 to 2 days; after
ammonium sulfate crystals are separated out, dialyzing and performing rotary
evaporation and concentration to small volume; finally, performing vacuum
freeze drying, thereby obtaining the sweet corn cob polysaccharide.
