Multifunctional core-shell structure fluorescent coding magnetic microspheres and preparation method thereof

The invention belongs to the technical field of nano materials and biomedicine, in particular to multifunctional core-shell structure fluorescent coding magnetic microspheres and a preparation method thereof. On the basis of ferroferric oxide nanoparticles synthesized by a hydrothermal process, pre-prepared coupled product of fluoresceins and amino propyl trimethoxysilane and ethyl orthosilicate are subjected to cohydrolysis in ammonia water to form a multifunctional fluorescent magnetic nano composite material with fluorescent and magnetic properties and high biological stability and biological adaptability. By regulating the mixing ratio of fluoresceins, namely fluorescein isothiocyanate (FITC) and rhodamine B isothiocyanate (RBITC), various fluorescent coding magnetic microspheres can be prepared, and the particle size of obtained material can be regulated according to the different ratios of the added ethyl orthosilicate to the ferroferric oxide. In addition, the obtained multifunctional composite nano material is subjected to amino silanization modification, so the biological application range of the novel fluorescent coding magnetic microspheres is further expanded and the novel fluorescent coding magnetic microspheres have bright application prospect in fields of biomedicine technology, medicine development, suspension chip and the like.

CRISPR/Cas9 targeted-knockout human TCAB1 gene and specificity gRNA thereof

The invention belongs to the technical field of the molecular biology and the biomedicine, and particularly relates to application of a gRNA sequence based on a CRISPR / Cas9 system to cause tumor cell apoptosis. According to the design principle of CRISPR / Cas9, two target points are designed on a human genome, and corresponding oligos is synthesized and established on a px458 carrier. The CRISPR / Cas9 system guided by the gRNA is established in a human hepatoma cell line (HepG2) according to the design of the two target points, a human TCAB1 gene can be effectively knocked out, the system is easy to operate, and the knockout efficiency of the human TCAB1 gene is high. The CRISPR / Cas9 system guided by the gRNA can be expected to be applied in novel tumor treating medicine.

Method for increasing non-homologous end joining efficiency of CRIPSR/Cas9 target knock-out genes

The invention belongs to the technical field of molecular biology and biomedicine, and relates to a method for increasing the efficiency of CRIPSR / Cas9 target knock-out genes NHEJ. According to the method for increasing the efficiency of CRIPSR / Cas9 target knock-out gene NHEJ, small molecule interference RNA that efficiently interferes with expression of PTEN genes is screened out; by using CRISPR / Cas9 system to target knockout genes, the small molecule RNA is cotransfected into the target gene, thus the NHEJ efficiency of target genes is effectively improved and the NHEJ of target gene in cell multiple targets is significantly improved. The method has the advantages of low cost, simple operation and high efficiency, and has a good promoting effect on the efficiency and application of CRISPR / Cas9 technology.

Elderly cognitive function classification method based on random forest

The invention relates to an elderly cognitive function classification method based on a random forest and belongs to the technical field of biomedicine. The method disclosed by the invention comprises the following steps: dividing the elderly cognitive function into three types by adopting MMSE scale scores and education levels; extracting a key cognitive domain influencing elderly cognitive function category classification by utilizing a cognitive function score relative ratio calculation method and a Pearson linearly dependent coefficient calculation method; establishing a random forest regression model, calculating attribute significance scores of non-scale attributes, and extracting external related attributes influencing the elderly cognitive function category classification; finally, equalizing the sample set based on the extracted key cognitive domain and external related attributes by adopting an SMOTE up-sampling method, and establishing an elderly cognitive function classification model by utilizing the random forest method. Compared with a scale classification method, the method provided by the invention has the advantages that the adopted attributes are few and easy to collect, and the method has high convenience; compared with other machine learning algorithms, subdivision of the elderly cognitive function types is realized, and research of a method for performing targeted intervention on the elderly cognitive functions is facilitated.

The invention belongs to the technical field of biomedicines, and in particular relates to application of seaweed polysaccharides. The seaweed polysaccharides can be taken as preparations for preparing anti-viral or immune medicines. Cell models and in-vivo animal experiments show that the seaweed polysaccharides can be used for significantly enhancing the animal immunity, and can also be used for promoting the generation of cell factors, the typing of T lymphocytes and the proliferation of mouse spleen cells so as to ensure that cellular immune reaction can be activated. The functional seaweed polysaccharides disclosed by the invention are natural polysaccharides which are extracted from seaweeds such as large seaweeds and kelps, gulfweeds, grateloupia sparsa, eucheuma muricatum, ulva pertusa kjellm, enteromorpha, gracilaria, ascophyllum nodosum and scytosiphon lomentaria, and can also be low-molecular-weight seaweed polysaccharides or seaweed oligosaccharides which are prepared by degrading the seaweed polysaccharides by virtue of different preparation methods. According to the application disclosed by the invention, a polysaccharide extract of single seaweed or a polysaccharide extract mixture of a variety of seaweeds can be taken as a novel anti-viral and immune enhancing agent to be applied to feeds of livestock, poultry, fish, shrimps and shellfish, and has wide application prospects.
