Synthesis and use of anti-reverse phosphorothioate analogs of the messenger RNA cap

New RNA cap analogs are disclosed containing one or more phosphorothioates groups. The analogs also contain modifications at the 2′-O position of 7-methylguanosine that prevent them from being incorporated in the reverse orientation during in vitro synthesis of mRNA and that hence are “anti-reverse cap analogs” (ARCAs). The ARCA modification ensures that the S atom is precisely positioned within the active sites of cap-binding proteins in both the translational and decapping machinery. The new S-ARCA analogs are resistant to in vivo decapping enzymes. Some S-ARCAs have a higher affinity for eIF4E than the corresponding analogs not containing a phosphorothioate group. When mRNAs containing the various S-ARCAs are introduced into cultured cells, some are translated as much as five-fold more efficiently than mRNAs synthesized with the conventional analog m7GpppG.

Accommodating intraocular lens with integral capsular bag ring

An intraocular lens (IOL) includes an optic for focusing light, an outer ring for supporting the optic in a capsular bag of an eye and a plurality of radially spaced apart, elongated intermediate members connecting the optic to the outer ring. The intermediate members are configured to convert radial forces exerted by the capsular bag on the support ring into axial movement of the optic, allowing a presbyopic patient to more effectively focus on near objects. The outer ring is preferably contoured to conform to the portion of the capsular bag between the anterior and posterior zonules, and has sufficient axial thickness to contact both sets of zonules. In addition, the edge of the ring includes at least one sharp edge corner to prevent epithelial cell growth toward the optic. In addition, the outer ring may include weakened areas configured to allow consistent and repeatable deformation in response to compressive forces.

Methods and systems for associating and translating virtual local area network (VLAN) tags

Methods and systems for associating and translating VLAN tags are disclosed. In one implementation, multiple different member VLAN tags are associated with a translation VLAN tag. When a frame addressed to an unknown MAC address and containing one of the member VLAN tags is received, the frame is flooded to the translation VLAN and to ports associated with the source member VLAN other than the originating port. In the copy sent to the translation VLAN, the VLAN tag is replaced with the tag of the translation VLAN, for tagged ports. For untagged ports, the input VLAN tag may simply be stripped from the frames. When a layer 2 frame addressed to an unknown MAC address and containing the translation VLAN tag is received, the layer 2 frame is flooded to each of the member VLANs and to ports of the translation VLAN other than the originating port. The VLAN tag in the frames forwarded to each of the member VLANs may be replaced with the VLAN tag corresponding to each member VLAN, for tagged ports. For untagged ports, the input VLAN tag may simply be stripped from the frames. MAC address learning for frames received from member VLANs extends to the translation VLAN but not to other member VLANs. MAC address learning for frames from the translation VLAN extends to the member VLANs.

The present invention relates to kits and methods for efficiently generating 5′ capped RNA having a modified cap nucleotide and for use of such modified-nucleotide-capped RNA molecules. The handle or actuator is configured such that movement of the handle is mimicked, not mirrored, by the end effector assembly. The mimicking motion can be achieved using various techniques, but in an exemplary embodiment the handle is located distal to an input joint, and motion is transferred to the end effector assembly through an output joint at a distal end of the elongate shaft. The motion is preferably transferred using a mechanical transmission coupled between the input and output joints.

In one embodiment, a device can include an outer tube. A rotatable shaft can be disposed within the outer tube can have a tissue harvesting tip formed on its distal end, the tissue harvesting tip being effective to excise tissue upon rotation thereof. In some embodiments, the tissue collection device can translate to indicate the amount of collected tissue. In many cases, devices disclosed herein can include driving mechanisms that are adapted to drive a tissue harvesting tip such that the tip excises soft tissue, but stops when contacting bone (or soon after contacting bone). In some embodiments, the tissue harvesting tip can be effective to excise viable tissue samples, such that the samples can exhibit desirable proportions of viable cells. Further, in some embodiments, the tissue harvesting tips can excise a tissue sample with tissue particles falling in certain size ranges.

The TPF-sDTF is bluntly dissected using the cryogenic probe such that a treating portion of the cryogenic probe is directly adjacent to a first treatment portion of the target nerve. The cryogenic probe is activated to create a first treatment zone at the first treatment portion of the target nerve to cause a therapeutic effect.
