The method comprises the following steps: firstly, preparing a quaternized polyethyleneimine and a hydroformylation polysaccharide cross-linking agent which have an anti-bacterial function; and then mixing and foaming the quaternized polyethyleneimine, the hydroformylation polysaccharide cross-linking agent and chitosan hydrochloride, uniformly coating the surface of a preprocessed gauze with foaming liquid, and carrying out freeze drying to obtain the antibacterial polysaccharide hemostatic cotton based gauze. After being compounded, polysaccharide hemostatic cotton and the preprocessed gauze are combined closely, do not fall off, have high intensity, and are suitable for compression hemostasis and binding at wounds. Moreover, compared with a common gauze, the antibacterial polysaccharide hemostatic cotton based gauze has the effects of inhibiting growth of microorganisms and inhibiting bacteria when stopping bleeding. In addition, a sponge coating the surface of the gauze uses safe and nontoxic polysaccharide as the cross-linking agent, so that the intensity and the blood stopping effect of the hemostatic cotton based gauze are further improved.
