The invention discloses a method for preparing beta-
chitin from the sector bone of a
squid. The method mainly overcomes the problems of severe environmental
pollution and great possibility of
chitin degradation in extraction of
chitin by using conventional acid-alkali methods. The method comprises the following steps:
drying and crushing the sector bone of a
squid, carrying out sieving, then adding EDTA decalcification solution and concentrated
hydrochloric acid, adjusting a pH value and carrying out ultrasonic water bath so as to obtain a decalcified reaction solution; adding acidic composite
protease into the reaction solution, carrying out ultrasonic-assisted
enzymatic hydrolysis and filtering and washing filter residue until the filter residue is neutral; and soaking the filter residue with a
hydrochloric acid solution and carrying out secondary decalcification and
drying so as to obtain a beta-chitin product. The method provided by the invention employs an ultrasonic-assisted EDTA process and
enzyme process to prepare beta-chitin and has the advantages of short time, simple operation, mild conditions, small
pollution to the environment and no influence on the structure of beta-chitin; and prepared beta-chitin has high purity and a good color.
