Carcinogenesis, also called oncogenesis or tumorigenesis, is the formation of a cancer, whereby normal cells are transformed into cancer cells. The process is characterized by changes at the cellular, genetic, and epigenetic levels and abnormal cell division. Cell division is a physiological process that occurs in almost all tissues and under a variety of circumstances. Normally the balance between proliferation and programmed cell death, in the form of apoptosis, is maintained to ensure the integrity of tissues and organs. Isolation And Use Of Solid Tumor Stem Cells

A small percentage of cells within an established solid tumor have the properties of stem cells. These solid tumor stem cells give rise both to more tumor stem cells and to the majority of cells in the tumor that have lost the capacity for extensive proliferation and the ability to give rise to new tumors. Thus, solid tumor heterogeneity reflects the presence of tumor cell progeny arising from a solid tumor stem cell. We have developed a xenograft model in which we have been able to establish tumors from primary tumors via injection of tumor cells in the mammary gland of severely immunodeficient mice. These xenograft assay have allowed us to do biological and molecular assays to characterize clonogenic solid tumor stem cells. We have also developed evidence that strongly implicates the Notch pathway, especially Notch 4, as playing a central pathway in carcinogenesis.

A plurality of electrodes are used to measure subepithelial impedance of parenchymal breast tissue of a patient at one or more locations and at least one frequency, particularly moderately high frequencies. The risk of developing breast cancer is assessed according to measured and expected or estimated values of subepithelial impedance for the patient and according to one or more experienced-based algorithms. Devices for practicing the disclosed methods are also provided.

Method and apparatus for measuring cancerous changes from reflectance spectral measurements obtained during endoscopic imaging

The present invention provides a new method and device for disease detection, more particularly cancer detection, from the analysis of diffuse reflectance spectra measured in vivo during endoscopic imaging. The measured diffuse reflectance spectra are analyzed using a specially developed light-transport model and numerical method to derive quantitative parameters related to tissue physiology and morphology. The method also corrects the effects of the specular reflection and the varying distance between endoscope tip and tissue surface on the clinical reflectance measurements. The model allows us to obtain the absorption coefficient ([mu]a) and further to derive the tissue micro-vascular blood volume fraction and the tissue blood oxygen saturation parameters. It also allows us to obtain the scattering coefficients ([mu]s and g) and further to derive the tissue micro-particles volume fraction and size distribution parameters.

Method and System for Detecting Electrophysiological Changes in Pre-Cancerous and Cancerous Tissue and Epithelium

Methods and apparatus are provided for determining the condition of selected regions of epithelial and stromal tissue. A plurality of electrodes are used to measure surface and transepithelial electropotential of tissue, especially breast tissue, as well as surface electropotential and impedance at one or more locations and at several defined frequencies, particularly low frequencies. An agent may be introduced into the region of tissue to enhance electrophysiological response characteristics. Measured and derived electrical properties facilitating diagnosis include DC potential, impedance, subepithelial resistance, epithelial resistance, capacitance, characteristic frequency, depression angle and transepithelial potential. Ambient and varying pressure measurements applied to epithelial tissue are also used for diagnosis. Improved diagnosis of tissue condition can be made based on the electrical properties and tissue electrophysiological response to altered ion transport and pressure. Devices for practicing the disclosed methods are also provided.
