CgHSP70 established plant expression vectors obtained by cloning from Zhongshan purple cinnamon of the chrysanthemum are transferred into the cut chrysanthemum by using an agrobacterium-mediated method for cultivation so as to initially obtain resistant plants, and positively transferred plants are obtained through the screening of hygromycin resistance; PCR (polymerase chain reaction) and fluorescence quantitative PCR analysis is performed on converted plants to prove that endogenous genes are transferred into genome DNA of transgenic plants and transcription occurs; and the resistance analysis on the offsprings of the transgenic plants proves that the resistance to high temperature, drought and high salt is obviously improved. In the invention, the stress resistance of the cut chrysanthemum is improved through the conversion of the endogenous CgHSP70 genes and normal transcription expression as well as the induction of the expression of the genes at an adverse environment; a novel and practical method is provided for selecting and breeding stress resistance varieties of chrysanthemum by using a gene engineering technology; and the breeding progress of the biotechnology of the chrysanthemum can be effectively accelerated.

The invention discloses an application of NsNHX1 protein and related biomaterials in breeding of reverse-resistant poplar. In the present invention, the nitraria sibirica Na<+> / H<+> reverse transport protein NsNHX1 is used as a research object, and is transferred into Populus tomentosa 84K to obtain transgenic NsNHX1 Populus tomentosa. Three transformants, N1, N2 and N3, are selected for identification of reverse resistance. The wild-type Populus tomentosa is used as a control to study the salt-tolerance and antioxidant capacity of transgenic NsNHX1 populus tomentosa under adversity stress. The results show that under the conditions of adversity stress treatment, the survival rate, biomass, leaf chlorophyll content, water content, plant height and antioxidant capacity of transgenic NsNHX1 are significantly higher than those of wild type populus tomentosa. The application indicates that NsNHX1 can significantly improve the reverse resistance and antioxidant capacity of the transgenic poplar, and NsNHX1 can be used as a salt tolerance gene to breed the reverse-resistant poplar varieties.

Treating system for industrial wastewater containing heavy metal and treating method thereof

The invention provides a treating system for industrial wastewater containing heavy metal. The system comprises an electrolytic tank (1); the electrolytic tank (1) comprises a first trough wall (11) and a second trough wall (12), which are arranged to be opposite; positive plates (2) and negative plates (3) are staggerly arranged in the electrolytic tank (1), the first ends of the positive plates(2) are fixed on the first trough wall (11), the first ends of the negative plates (3) are fixed on the second trough wall (12), and each negative plate (3) is a starting sheet of a tri-dimensional network structure. According to the invention, because of utilization of the tri-dimensional network structure in the treating system, the negative starting sheets have substantially improved capacity in bearing stress, and the problem of short circuit of cathodes and anodes caused by cracking of the negative plates is avoided; the operation of plate stripping is omitted. The invention also provides a treating method for industrial wastewater containing heavy metal by using the treating system.

The amino acid sequence of the protein is shown in a sequence 1 of a sequence table and can be used for regulating and controlling the stress tolerance of the plants, or substances and methods, capable of adjusting the expression level of the protein shown in the sequence 1 of the sequence table, can be used for regulating and controlling the stress tolerance of the plants. Shown by experiments, after the coding gene of the protein AtbHLH122, which is shown in a sequence 2 of the sequence table, is over-expressed in Arabidopsis thaliana, a transgenic Arabidopsis thaliana plant is obtained, the survival rate after drought treatment, the germination rate and leafy rate after 140mM NaCl treatment and the root length after 250mM mannitol treatment of the transgenic Arabidopsis thaliana plant are remarkably lower than those of the wild plants, and furthermore, after the treatments, gene AtbHLH122 knockout mutants are less than those of the wild plants; and thus, the protein AtbHLH122 shown in the sequence 1 of the sequence table has the function of regulating and controlling the stress tolerance of the plants and has an important significance on the aspects of stress tolerance research and breeding of the plants.
