Viable dried bacteria produced by drying in the presence of trehalose and divalent cation

A method is provided for preserving live bacteria by subjecting an aqueous system containing the growing bacteria to drying without special equipment, in the presence of trehalose with or without the addition of divalent cations as stabilizing agents. Further, a dried composition for preservation of aerobic bacteria in a viable state is provided. The dried composition consists essentially of dried viable aerobic bacteria and an appropriate growth medium. The bacteria and growth medium are initially placed in an aqueous solution of 10 mM to 200 mM trehalose and a divalent cation, and dried at room temperature.

Systems and methods for cell preservation

The present invention generally relates to devices and methods for the preservation of cells using drying, freezing, and other related techniques. In one set of embodiments, the invention allows for the preservation of cells in a dried state. In another set of embodiments, the invention allows for the preservation of cells within a glass or other non-viscous, non-frozen media. In some embodiments, the invention allows for the preservation of cells at temperatures below the freezing point of water, and in some cases at cryogenic temperatures, without inducing ice formation. The cells, in certain embodiments, may be preserved in the presence of intracellular and / or extracellular carbohydrates (which may be the same or different), for example, trehalose and sucrose. Carbohydrates may be transported intracellularly by any suitable technique, for example, using microinjection, or through non-microinjected methods such as through pore-forming proteins, electroporation, heat shock, etc. In certain instances, the glass transition temperature of the cells may be raised, e.g., by transporting a carbohydrate intracellularly. In some cases, the cells may be dried and / or stored, for example, in a substantially moisture-saturated environment or a desiccating environment. The cells may also be stored in a vacuum or a partial vacuum. The cells may be protected from oxygen, moisture, and / or light during storage. In certain cases, an inhibitor, such as a cell death inhibitor, a protease inhibitor, an apoptosis inhibitor, and / or an oxidative stress inhibitor may be used during preservation of the cells. The cells may be stored for any length of time, then recovered to a viable state, e.g., through rehydration, for further use.

Method for preparing exosome freeze-dried powder of human amniotic mesenchymal stem cells

The invention relates to the field of stem cells, discloses a method for preparing exosome freeze-dried powder of stem cells, and particularly relates to a method for preparing exosome freeze-dried powder of human amniotic mesenchymal stem cells. The method for preparing the exosome freeze-dried powder of the human amniotic mesenchymal stem cells disclosed by the invention comprises the following steps: mixing trehalose with exosomes of the human amniotic mesenchymal stem cells as a cryoprotectant; filtering and removing bacteria by virtue of a filter membrane; and firstly carrying out ultralow temperature pre-freezing, and then carrying out low-temperature freezing at a certain vacuum degree. The preparation method disclosed by the invention is simple to operate, safe and effective. An experiment proves that compared with a conventional preparation method of the freeze-dried powder, the morphologies and the activity of the exosomes of the human amniotic mesenchymal stem cells can be well kept by the method for preparing the exosome freeze-dried powder of the human amniotic mesenchymal stem cells disclosed by the invention; and the method is suitable for long-term preservation and application of the exosomes of the human amniotic mesenchymal stem cells.

Hydrotreating carbohydrates

Conversion of renewable hydrocarbons to transportation fuels is required to reduce carbon emission, limit the use of fossil fuels, and develop renewable energy sources. Sorbitol, xylitol and trehelose are polyalcohols generated from the liquefaction of various sugars and carbohydrates in biomass from algae, corn, sugarcane, switchgrasses, and biological wastes. Mixtures of aqueous polyols and fuel feedstocks are catalyzed over metal catalysts to produce hexanes, pentanes, and lighter hydrocarbons. By managing the catalyst, reaction conditions and sulfur content, the octane value of the product fuel is dramatically increased.
