The invention discloses a temperature response type core cross-linked star polymer with a glycoprotein enriching effect and a preparation method thereof. According to the synthesized temperature response type core cross-linked star polymer with the glycoprotein enriching effect, an outer arm is prepared from a recognizing unit, i.e., phenylboronic acid, which plays a role in recognizing glycoproteins, and can be specifically bound to the glycoproteins, so as to complete a glycoprotein trapping process; the cross-linked core is prepared from a temperature response type polymer and can subject to a swelling 'dissolving'-precipitating change along with temperature changes, so as to complete a process of separating the glycoproteins from a mixed solution. The method has the advantages that the influence caused by interfacial mass transfer in an enriching process is eliminated, the capacity of enrichment is increased greatly, and the time for incubation is shortened; meanwhile, compared with the other methods, recognizing units are fixed by an arm precedence method, so that the operation is relatively simple, the fixed amount is high, and thus, the capacity of enrichment is further increased. In addition, the method can achieve the effective and rapid separation of the glycoproteins through simple centrifugation or filtration, so that the requirements on modern bioseparation are met.
