The invention discloses a
volvariella volvacea spore mutagenesis method which comprises the following steps of (1) collecting
volvariella volvacea spores; (2) scraping the spores into
sterile water bya small sterile knife, performing
dilution, and stopping
dilution when the
spore concentration reaches 10<6> to 10 <8> / mL; (3) before working, firstly starting a cooling
water circulation machine, then opening a
helium gas valve, enabling the gas input pressure to be between 0.15 MP and 0.2 MP, performing
ultraviolet sterilization for 20 min, firing sterilized slide glass for 30s on an
alcohol lamp, and putting the fired sterilized slide glass into a sterile empty plate; and (4) taking 1 mL of a
spore suspension, adding 143 mL of sterile
glycerol with the concentration being 5 percent, performing uniform shaking, recording the liquid as mother liquid, sucking 10 muL of the mother liquid, dripping the mother liquid onto the slide glass in a suspended way, transferring the sterile plate into an ARTP
operation room, taking and putting the slide glass onto a corresponding
mutagenesis position by a pair of special sterile
tweezers, clamping an EP tube containing 1 mL of
sterile water underthe
mutagenesis position, and performing mutagenesis under the condition of the input power being 100 W, the
jet flow being 10 SLW and the
irradiation mutagenesis distance being 2 mm. The volvariellavolvacea spore mutagenesis method has the advantages that the back
mutation probability caused by passage can be fundamentally reduced.
