The method comprises the steps that paper mulberry branches, sweet potato vines, rotten wood, sweet potato powder and bagasse are mixed, smashed and screened, then water is added in a stirring process, natural fermentation is conducted, tea powder, gypsum powder, lime powder, calcium carbonate, glucose and magnesium sulfate are added, and uniform mixing is performed to obtain a cultivation material; the cultivation material is put into a fungus bag, high-pressure sterilization is performed, natural cooling is performed to the room temperature, and the fungus bag is taken out from a pot to obtain a ganoderma lucidum bag material; after the ganoderma lucidum bag material is inoculated, thetemperature is controlled to be 24-26 DEG C in the first 10-12 days; then the temperature is adjusted to 21-23 DEG C, the temperature of 21-23 DEG C is maintained to the 15th-20th days, and then the temperature is adjusted to 20-21 DEG C; in a mycelial growth period, when mycelia grow to be 14-16 cm, the fungus bag is removed; and after the mycelia are fully distributed, the mycelia are moved intoa ganoderma lucidum output greenhouse and placed on a three-dimensional cultivation fungus frame, after arrangement is completed, each layer is covered with a nutrient medium, the humidity in the ganoderma lucidum output greenhouse keeps 80-88%, and harvesting is conducted after fungus caps grow until white spots disappear.
