Novel synthetic binding pairs and uses thereof

Derivatized cucurbiturils can be efficiently prepared and utilized for forming cucurbituril assemblies. These cucurbituril assemblies, in turn, can form affinity binding pairs with polyamines structures that are designed capable of binding to these assemblies. The resulting affinity pairs are highly advantageous over the presently known affinity pairs and therefore can be efficiently utilized in a myriad of application. Thus, according to one aspect of the present invention there is provided a cucurbituril comprising at least one functional group covalently attached hereto, whereby the at least one functional group being for forming an assembly of at least two cucurbiturils. According to further features in preferred embodiments of the invention described below, the cucurbituril is selected from the group consisting of CB[5], CB[6], CB[7], CB[8] and CB[n], wherein n is an integer that equals to or is lower than 20. Preferably the cucurbituril is selected from the group consisting of CB[5], CB[6], CB[7] and CB[8]. According to still further features in the described preferred embodiments the assembly is selected from the group consisting of a dimer, a trimer, a polymer, an oligomer, a dendrimer and a cluster of the cucurbituril.

The invention provides a seven-membered cucurbituril-rare earth metal linear tubular supramolecular polymer and a synthetic method. When cadmium chloride exists, the seven-membered cucurbituril-rare earth metal linear tubular supramolecular polymer is formed by seven-membered cucurbituril (Cucurbit [7] uril and Q [7]) and rare-earth metal salt (Ln (NO3)3 or LnCI3) in 6M hydrochloric acid solution. According to the seven-membered cucurbituril, dosing mole ratio of the rare-earth metal salt to the cadmium chloride ranges from 1:6 to 8:2-8, and mixture of the rare-earth metal salt and the cadmium chloride are heated and mixed evenly and stand for several days under normal temperature to form a target compound. The method has the advantages of being simple in the synthetic method, easy and simple to operate, high in productivity (between 60% and 80%), short in period and the like. Compared to pure seven-membered cucurbituril, the seven-membered cucurbituril-rare earth metal linear tubularsupramolecular polymer synthetized by the method has strong adsorption to methyl alcohol, ethanol and acetone.

The invention relates to a supermolecular assembly for small interfering RNA targeting delivery. The supermolecular assembly is a four-element supermolecular assembly synthesized on the basis of beta-cyclodextrin modified hyaluronic acid, an adamantine polyamine compound, cucurbituril [6] and small interfering RNA. The four-element supermolecular assembly is based on the strong non-covalent interaction between beta-cyclodextrin and adamantine and between adamantine polyamine chain and cucurbituril [6], and the electrostatic interaction between the adamantine polyamine chain and small interfering RNA, and forms supermolecular nanoparticles with hydrophilic hyaluronic acid as the shell. The supermolecular assembly provided by the invention has the advantages of simple synthesis and construction route, low production cost and high yield, and is suitable for amplified synthesis and actual production application. The method comprises the following steps: carrying out a mixed reaction on a glycoluril-aldehyde polymer, an acidic ionic liquid and a solvent or carrying out the mixed reaction on glycoluril, aldehyde, the acidic ionic liquid and the solvent for 1 second to 300 hours at a reaction temperature of 0 DEG C to 300 DEG C; and standing a reaction liquid and cooling after the reaction, thereby separating out the cucurbituril or the cucurbituril derivative in a crystallization manner; or adding an organic solvent after the cooling of the reaction liquid, thereby separating out the cucurbituril or the cucurbituril derivative in a precipitation manner. Compared with the background technology which is strong in corrosivity, serious in acid pollution and low in yield, the synthesis process provided by the invention is environmental friendly, economical and efficient, and has the advantages that the acidic ionic liquid is used for replacing the traditional inorganic strong acid to serve as a catalyst, thereby being environmental friendly; the acidic ionic liquid can be recycled, thereby being economical; the microwave radiation is optimally adopted as a heat source, thereby being efficient.
