The invention relates to a method for preparing cellulose microfibril and a film thereof from wheat straw. The method comprises the following steps: screening wheat straw fiber slurry; carrying out enzyme hydrolysis bleaching on wheat straw fibers; carrying out high-pressure homogenization treatment and preparing cellulose microfibrils; and preparing the cellulose microfibril film. According to the preparation method, the bleached wheat straw fiber is taken as a raw material, and the cellulose microfibril is prepared by combining enzyme pretreatment with a high-pressure homogenization method, so that the surface morphology and pore structure of the cell wall of the wheat straw fiber are changed, the preparation process of the cellulose microfibril is further optimized, and the improvement of hydrophilicity, mechanical properties, transparency and other properties of the cellulose microfibril is promoted; therefore, the cellulose microfibril (CMF) film with ideal performance is prepared by a low-energy-consumption and low-cost method, further market development and utilization of the cellulose microfibril (CMF) are greatly promoted, and agricultural comprehensive utilization benefits are improved.
