Culture medium capable of simultaneously enriching five kinds of food-borne pathogenic bacteria and preparation method for culture medium

The invention relates to a culture medium for simultaneous composite enrichment of five kinds of food-borne pathogenic bacteria, namely Salmonella, Escherichia coli, Staphylococcus aureus, Listeria monocytogenes and Shigella, and a preparation method for the culture medium. Food-borne pathogenic bacteria are a significant reason to cause food positioning, so the rapid and accurate detection of the food-borne pathogenic bacteria has an important practical significance for preventing and controlling food safety incidents. The culture medium is characterized by comprising the following components: 10.0 g of peptone, 10.0 g of sodium chloride, 9.0 g of disodium hydrogen phosphate, 1.5 g of monopotassium phosphate, 0.1 g of cholate, 0.1 mg of potassium tellurite, 1.0 g of lithium chloride, 3.0 g of glucose, 2.0 g of mannitol, 2.5 g of sodium pyruvate, 1.0 g of aesculin and 1,000 mL of distilled water, wherein the pH value is 7.1 to 7.5. The culture medium can simultaneously enrich five kinds of target pathogenic bacteria, can be used for separation and identification of target bacteria, can also be used for the molecular detection of multiple pathogenic bacteria on the same detection platform, provides technical support for a method for rapidly detecting five kinds of pathogenic bacteria in food, and meets the requirement of simultaneous detection of five kinds of food-borne pathogenic bacteria.

Primers and method of detecting bacteria

The first primer of the invention is a primer which, when used in PCR under appropriate conditions, serves to detectably amplify 16S rRNA-encoding DNAs of bacteria of the Escherichia, Salmonella and Vibrio genera, but when used in PCR under the same conditions, does not serve to detectably amplify either chloroplast 16S rRNA-encoding DNAs or mitochondrial 16S rRNA-encoding DNAs. The second primer of the invention is a primer which, when used in PCR under appropriate conditions, serves to detectably amplify 16S rRNA-encoding DNAs of Staphylococcus aureus and Bacillus cereus, but when used in PCR under the same conditions, does not serve to detectably amplify either chloroplast 16S rRNA-encoding DNAs or mitochondrial 16S rRNA-encoding DNAs.

Multiple rapid detection method for three food borne pathogenic bacteria, and detection primer set and kit thereof

The invention discloses a multiple rapid detection method for three food borne pathogenic bacteria, and detection primer sets and a detection kit thereof. An upstream primer and a downstream primer of a rapid detection primer set for Salmonella have sequences as represented by SEQ No.1 and SEQ No.2, respectively; an upstream primer and a downstream primer of a rapid detection primer set for Shigella have sequences as represented by SEQ No.3 and SEQ No.4, respectively; an upstream primer and a downstream primer of a rapid detection primer set for o Staphylococcus aureus have sequences as represented by SEQ No.5 and SEQ No.6, respectively. Bacterial genome DNA extracted from a sample to be tested is subjected to a LAMP reaction with the primer sets in a same reaction system, and the resultant of the reaction is identified to determine whether the sample contains Salmonella, Shigella or Staphylococcus aureus. Every primer set provided in the invention has strong specificity and can accurately detect genome DNAs of Salmonella, Shigella and Staphylococcus aureus in a same reaction system.

Lactobacillus plantarum SWUN5815 for high-yield bacteriocin and application of lactobacillus plantarum SWUN5815

The invention discloses a lactobacillus plantarum SWUN5815 for high-yield bacteriocin and an application of the lactobacillus plantarum SWUN5815. The strain is preserved at the China Center for Type Culture Collection (CCCTCC) on August 2, 2016 and the preservation number is CCTCC NO: M2016419; and the strain is separated from yak dung. The bacteria generated by the strain can be degraded by protease; the strain has a very high inhibition effect on salmonella and escherichia coli and has good acid resistance and relatively high cholate tolerance; the survival rate after 3h in artificial gastric juice of which the pH is 3.0 is 93.36+ / -4.06%; the strain can slowly grow in 1.0% cholate; and the growth efficiency reaches 15.26+ / -1.19% of that of the strain which is cultivated under the condition free of the cholate. The bacteriocin is capable of effectively inhibiting growth of salmonella, escherichia coli and staphylococcus aureus. Therefore, the lactobacillus plantarum SWUN5815 for the high-yield bacteriocin has the potential of being applied to yak intestinal tracts as a microecological preparation.

Preparation method of bacteriophage lyase capable of lysing escherichia coli and salmonella

Belonging to the field of animal biomedical engineering, the invention specifically discloses a preparation method of a bacteriophage lyase capable of lysing escherichia coli and salmonella. Through primer design, a lyase gene of bacteriophage ECGD1 is successfully amplified by PCR, and is transferred into an expression system to obtain a recombinant strain. IPTG induction is carried out on the recombinant strain to obtain expression, then the strain is lysed to obtain a soluble recombinant lyase of bacteriophage ECGD1, the recombinant lyase can lyse a plurality of escherichia coli and salmonella, i.e. having a wide lysis spectrum. In addition, the synergistic effect result of the recombinant lyase and polymyxin B shows that the existence of the recombinant lyase lowers the minimum inhibitory concentration of polymyxin B, i.e. the two have a synergistic bacteriostatic effect. Identification on the lysis activity of the recombinant lyase and identification on the wide lysis spectrum thereof reveal that the recombinant lyase of the bacteriophage ECGD1 has potential application value in prevention and control of escherichia coli and salmonella infection.
