The invention provides a method for extracting
Humulus scandens (Lour.) Merr. leaf
protein and total flavonoids in a graded manner simultaneously from fresh and tender stems and leaves of
Humulus scandens (Lour.) Merr.. The method comprises the steps as follows: firstly, the stems and the leaves of the
Humulus scandens (Lour.) Merr. are obtained, the
water content and the
wet weight are measured after the stems and the leaves are washed and dried by airing, and the stems and the leaves are
cut into slices; then
distilled water is added for homogenization, press
filtration is performed after homogenization to obtain Humulus scandens (Lour.) Merr. juice and filter cakes; the Humulus scandens (Lour.) Merr. juice is subjected to heating, alkaline
precipitation and
centrifugation to obtain a supernatant liquid and deposits, the deposits are frozen and dried so as to obtain the Humulus scandens (Lour.) Merr. leaf
protein; then the filter cakes and the supernatant liquid are mixed, extraction is performed under an alkaline condition, acid
precipitation and
centrifugation are performed, and after obtained deposits are washed by water, dissolved in
alcohol and subjected to suction
filtration, a filtrate is concentrated, dried and frozen so as to obtain the Humulus scandens (Lour.) Merr. total flavonoids. According to the method, the Humulus scandens (Lour.) Merr. leaf
protein and the Humulus scandens (Lour.) Merr. total flavonoids are extracted in the graded manner simultaneously in one technological process, so that the method is convenient to operate, accurate and reliable in extraction technological condition and high in yield and purity, two important effective components, namely, the leaf protein and the total flavonoids, in the Humulus scandens (Lour.) Merr. can be developed and utilized simultaneously, and development and utilization values of Humulus scandens (Lour.) Merr. resources are increased.
