water sample, which comprises the following steps: firstly, material cleaning: cleaning a glass ware, rubber plugs and
aluminium foil covers; secondly, preparing a NR-SOB testing culture medium; thirdly, sterilization: accommodating the prepared NR-SOB testing culture medium into the glass ware, sealing by a gauze, and using a
steam sterilization pot to sterilize; fourthly,
deoxidization,
sulfur treatment and split charging:
deoxidization: placing the culture medium after sterilization into an anaerobic
workstation for
deoxidization,
sulfur treatment: adding a Na2S.9H2O
mother liquor into the culture medium, and split charging: using a liquid-moving
machine to accommodate the culture medium into cleaned glass bottles respectively; and fifthly, NR-SOB detection: adopting an extinct
dilution method to detect the number of
bacteria in a
water sample to be detected. The detection method has the advantages of simplicity in operation, convenience for preparation, convenience and quickness for testing, accurate data, high reliability, economy and practicability, quickness, accuracy, convenience for microbial detection in an oilfield and the like.
