The invention discloses a forced nucleus retention method in nucleated pearl cultivation and a nucleus retainer used in the method. The method includes: fastening the base to the edge of the pearl shell through bolts or fastening the base to the outer side of the pearl shell in an adhered manner, extending one end of the nucleus retaining bar into an outer holding cavity of the pearl shell, exerting certain pressure to hold the nucleus retaining bar onto the outer side of a visceral sac at the position of the nucleus, and fastening the other end of the nucleus retaining bar onto the base; placing the pearl shell in culture water for 10 to 15 days of temporary culture, and removing the nucleus retainer; and forcedly retaining the nucleus which is implanted and has a wound healed in the pearl shell without 'exposure'. The forced nucleus retention method and the nucleus retainer are effective in nucleus fixation, nucleus retention rate of nucleus implanted shells can reach 100% under experimental conditions, and the problem that a nucleus of a large nucleated pearl is 'exposed' during culturing is solved.
