Method for identifying pathotype of verticillium dahlia, kit and contained nucleic acid and primer pair

The invention provides nucleic acid which is provided with a sequence as shown in SEQ ID NO:1. The invention further provides a primer pair for amplifying the nucleic acid and a method for identifying a pathotype of a fungi strain. The fungi strain is verticillium dahlia. The method comprises the following steps: (1) preparing a sample to be tested, containing whole genome DNA (deoxyribonucleic acid) of the verticillium dahlia; (2) taking the sample to be tested, containing whole genome DNA obtained from the step (1) as a template and carrying out PCR (polymerase chain reaction) amplification by the primer pair; detecting if nucleic acid as shown in SEQ ID NO:4 is existent in an amplified product; (3) judging the pathotype of the verticillium dahlia according to a detection result in the step (2), and when nucleic acid as shown in SEQ ID NO:4 is inexistent in the amplified product, indicating that the pathotype of the verticillium dahlia belongs to a high pathotype; when nucleic acid as shown in SEQ ID NO:4 is existent in the amplified product, indicating that the pathotype of the verticillium dahlia does not belong to the high pathotype.

Application of ambrosia artemisiifolia essential oil as verticillium dahliae bactericide

The invention relates to application of ambrosia artemisiifolia essential oil as a verticillium dahliae bactericide. Growth of verticillium dahliae is inhibited by a method of enabling the verticillium dahlia to be in contact with the ambrosia artemisiifolia essential oil and fumigating the verticillium dahlia with the ambrosia artemisiifolia essential oil. Results show that the ambrosia artemisiifolia essential oil has a significant effect on mycelial growth of the verticillium dahliae at a low concentration of 0.5 mu l / ml, and the growth inhibition effect is improved gradually with the increase of the concentration; when the concentration is continuously increased to be 5 mu l / ml, the mycelia of inoculated stems all lose the activity of continuous expansion, and the growth of the myceliais completely inhibited; in a fumigation experiment, the ambrosia artemisiifolia essential oil with the concentration of 0.2 mu l / ml fumigates the stems of the verticillium dahlia, and the mycelia ofthe stems are significantly inhibited; and when the concentration reaches 1.2 mu l / ml, the mycelia are killed completely to lose activity. The growth of the verticillium dahliae is inhibited by the ambrosia artemisiifolia essential oil provided by the invention, the ambrosia artemisiifolia essential oil is derived from plants, can be decomposed in nature, is environmentally friendly and does notpollute the environment, and meanwhile, waste harmful plants are utilized.
