The invention discloses preparation and application of total alkaloids extract and effective components in peganum plant seeds. The peganum plant (comprising harmel, multisectum peganum and peganum nigellastrum) seeds are heated, refluxed and extracted by ethanol, dissolved along with heat, acidulated by added 2 to 10 percent hydrochloric acid, and basified by ammonia water to separate out sediment; and the sediment is dried to obtain the total alkaloids extract mainly containing dehydrogenized peganine and peganine according to a proportion of between 0.18 to 1 and 4.30 to 1, wherein the content of the total alkaloids is more than 50 percent. The total alkaloids extract is separated by chromatography to obtain ten effective monomer components such as the dehydrogenized peganine, the peganine, and the like. Through thin layer chromatography and biological self-developing analysis, the total alkaloids extract and the effective monomer components thereof have the effect of resisting activity of acetylcholine esterase, and can be used for preparing medicament for resisting the activity of the acetylcholine esterase and medicament for treating neurodegenerative diseases.

Enzyme inhibition method for detecting carbamates

The invention relates to the chemical field and in particular to an enzyme inhibition method for detecting carbamates. Ethyl carbamate is oxidized by using an oxidant in the presence of an acetylcholine iodide substrate, a 5,5'-dithio bis-(2-nitrobenzoic acid) (DTNB) developer and an acetylcholin esterase with the activity of more than 200U / g, and then the absorbance variation value of the ethyl carbamate along with time at 3min is detected by spectrophotometry to obtain the inhibition condition of the ethyl carbamate on the acetylcholin esterase so as to obtain the content of the ethyl carbamate. The substrate and the developer used in the invention are low in cost; the inhibition rate is greatly increased since the ethyl carbamate is oxidized by using the oxidant; and the method is rapid, simple, high in sensitivity, mild and easily controlled in reaction conditions, simple in steps and easy to operate and needs less equipment. According to the principle of the method, a rapid kit is developed for the rapid detection of ethyl carbamates in alcoholic beverages and fermented foods.

Preparation for detecting organophosphorus pesticide residual test paper and its use method

The present invention discloses a preparation method of a test paper for testing the residual organphosphorous pesticide and a using method thereof, wherein the preparation method comprises the following procedures: (1) preparing acetylcholine esterase with fresh blood of hen or female duck as raw material; (2) fixing the acetylcholine esterase on the filter paper; (3) slicing when the filter paper is frozen and dried. A substrate liquid, a color developing liquid and a sensitization liquid are provided. The method of the invention has the advantages of low cost, simple and fast operation. The invention discloses a series of 11-replaced oxoisoaporphine derivatives as well as a synthetic method and an application thereof. The synthetic method comprises the following steps: (1) carrying out ring closing reaction on 3-chlorophthalic anhydride and phenylethylamine as raw materials so as to construct a 10-Cl-1-azabenzanthrone parent body; (2) nitrating the parent body compound so as to obtain a 11-site nitrated product, and reducing the 11-site nitrated product so as to obtain 11-amino-10-chlorine-7H-dibenzoquinoline-7-ketone; and (3) reacting the 11-amino-10-chlorine-7H-dibenzoquinoline-7-ketone with an acyl chloride compound connected with piperidine so as to obtain a corresponding target product. Through study, the applicant finds that the series of derivatives have very strong inhibitory activity on acetylcholin esterase and are expected to be used for treating AD (Alzheimer Disease), cerebrovascular dementia and related diseases caused by cholinergic neurotransmitter reduction.
