The invention discloses a segmented
plant gene sequence. According to the segmented
plant gene sequence, a
cadmium (Cd) strongly-induced rice specific
gene promoter CdS2 is separated, cloned and identified from
Oryza sativa L cv.Nipponbare, and acquired transgenic rice is subjected to
cadmium (Cd) treatment and is subjected to GUS chemical
dyeing to find that each part of treated transgenic plants is significantly dyed; and moreover, by analyzing Gus
gene expression quantities before and after RT-qPCR identification treatment, results show that the Gus
gene expression quantity of the transgenic plants after being subjected to
cadmium (Cd) treatment is 887.9 times that of the transgenic plants before being subjected to cadmium (Cd) treatment and can reach 4 times of
ACTIN activity of a constitutive
promoter. The
promoter disclosed by the invention can be used for reflecting the
cadmium pollution strength in soil according to the expression quantity of the promoter in rice, has the characteristics of simplicity in operation, low
detection limit, micro sensitivity and high accuracy, can play important significance in researching environment relationships, improving ecological environments and achieving
sustainable development, can also be used for actively analyzing
pollution formation principles, and ensures that valuable experiences for avoidance and controllable transformation of
cadmium pollution can be further provided.
