The invention relates to a guided tissue regeneration membrane, which comprises a compact coating and a loose coating, wherein the compact coating is composed of collagen-I or collagen-I composite hyaluronic acid; and the loose coating is composed of collagen-I or collagen-I composite calcium salt. A preparation method of the guided tissue regeneration membrane comprises the following steps of: preparing a collagen solution or a collagen and hyaluronic acid mixed solution by the use of acetic acid; injecting the solution into a self-made die, and drying it in the air to prepare the compact coating; injecting the collagen solution or a mixed liquor of the collagen solution and calcium salt onto the compact coating, and uniformly spreading; precooling the die at ultralow temperature, followed by freeze-drying to prepare the guided membrane with the compact coating and loose coating structure; carrying out vacuum high-temperature crosslinking and chemical crosslinking; and finally cleaning. The regeneration membrane has good histocompatibility and mechanical strength, low antigenicity and strong guided tissue regeneration capability. Its degradation rate in vivo is 3-8 months. The method is easy to operate, is suitable for automatic and continuous large-scale production, and solves the problems of large size of collagen membrane and material uniformity during industrial production.

Collagen membranes are formed from porcine rinds (i.e., pig skins) for us in a variety of applications and, most preferably, for wrapping food products, such as hams and the like. First, after removing skins from the porcine, the skins are promptly frozen. In later processing, the rinds are thawed and then enzymatically defatted. Then, a quick alkalinic hydrolyzation is performed on the rinds. Then, an acidic hydrolyzation is performed on the rinds. The rinds are then ground into a gel-like fluid mass. Finally, the fluid mass is extruded, sheeted and dried into a collagen membrane. The collagen membrane produced can be, in preferred embodiments, used for wrapping food products, such as hams.

The method comprises: a pretreatment step, i.e. collecting a mammalian peritoneum, conducting flowing water washing, hair removal and preliminary degreasing for standby use; an antigen removal treatment step, i.e. carrying out further degreasing on the standby peritoneum by means of an enzyme treatment process, an alkali treatment process, an acid treatment process and a hypertonic treatment process, and removing immunogenicity; a collagen fiber contraction treatment step, i.e. carrying out dehydration, non-hydrophilic substance removal and washing treatment on the peritoneum subjected to the antigen removal treatment so as to contract the collagen fibers of the peritoneum; and a drying step, spreading the contracted peritoneum on a culture dish surface to make the compact layer of the peritoneum contact the culture dish, rapidly transferring the peritoneum into a refrigerator at a temperature ranging from -60 to -80DEG C to undergo pre-freezing for 1-8h, then freeze-drying the peritoneums in a freeze dryer for 24-36h, thus obtaining a collagen membrane able to guide tissue regeneration. The guided membrane provided by the invention has a compact layer and a loose layer, also has good biocompatibility, hydrophilicity and bone induction ability, and has a long in-vivo degradation time.
