Thermal cycler for PCR

An instrument for performing highly accurate PCR employing an assembly, a heated cover and an internal computer. The assembly is made up of a sample block, a number of Peltier thermal electric devices and heat sink, clamped together. The sample block temperature is changed exclusively by the thermoelectric devices controlled by the computer. The sample block is of low thermal mass and is constructed of silver. The Peltier devices are designed to provide fast temperature excursions over a wide range. The heat sink has a perimeter trench to minimize edge losses and is adjacent to a continuously variable fan. A perimeter heater is used to improve the thermal uniformity across the sample block to approximately ±0.2° C. A heated platen pushes down onto the tube caps to apply a minimum acceptable force for seating the tubes into the block, ensuring good thermal contact with the block. The force is applied about the periphery of the tube caps to prevent distortion of the caps during thermal cycling. The platen is heated to provided thermal isolation from ambient conditions and to prevent evaporation from the surface of the sample into the upper portion of the sample tube. A control algorithm manipulates the current supplied to the thermoelectric coolers such that the dynamic thermal performance of the block can be controlled so that pre-defined thermal profiles for the sample temperature can be executed. The sample temperature is calculated instead of measured using a design specific model and equations. The control software includes calibration diagnostics which permit variation in the performance of thermoelectric coolers from instrument to instrument to be compensated for such that all instruments perform identically. The block / heat sink assembly can be changed to another of the same or different design. The assembly carries the necessary information required to characterize its own performance in an on-board memory device, allowing the assembly to be interchangeable among instruments while retaining its precision operating characteristics. The instrument has a graphical user interface. The instrument monitors the thermoelectric devices and warns of changes in resistance that may result in failure.

A fingerprint-reading system including a sensor on which a finger, whose imprint is to be read, is pressed. The sensor has an active surface of elements sensitive to variations in temperature and integrated heating resistors that create a transient variation in the temperature of the sensitive elements thereby to generate temporary charges in each element when pressed by a finger. The charges are output in synchronism with creation of the transient variations in the temperature of the sensitive elements to provide stable images of the fingerprint which may be motionless. The output of the sensor enables real-time display of the fingerprint images and processing of authentication data. The heating resistor enables the sensor to produce stable images of a fingerprint by producing an internal excitation to offset the tendency of electrical charges, induced by variations of the physical temperature of the finger, from disappearing because the temperature of the finger has reached equilibrium.

Method and system for determining atmospheric profiles using a physical retrieval algorithm

The present invention provides a new approach for processing hyper-spectral radiance data. It uses a transformation matrix to convert an instrument radiance spectrum into a pseudo-monochromatic radiance spectrum. The pseudo-monochromatic radiance spectrum is produced by an empirical transform of the instrument channel spectrum to a monochromatic equivalent spectrum (i.e., a pseudo-monochromatic spectrum). Eigenvector regression is used to produce the empirical transformation. Although the transformation does not produce the monochromatic radiance spectrum without error, the transformation error is generally well below nominal instrument noise levels for most spectral channels. The reduction in instrument noise results from a noise filtering effect of the eigenvector transformation. One of the advantages of the present invention is that it eliminates the need to build different fast radiative transfer models (RTMs) for different observing instruments, since the retrieval of geophysical parameters is based on an inversion of the monochromatic radiative transfer model. Although a different transformation matrix is required for different instrument spectral channel characteristics, the production of this transformation matrix is straightforward and simpler than the production of an accurate channel radiance fast model.

Temperature control using infrared sensing

A system and method for sensing and controlling the temperature of a heating, ventilation and air conditioning (HVAC) system utilizes an infrared sensor. The infrared sensor is used to detect temperature over several different areas on an evaporator surface on which a cold spot may occur. More preferably, the infrared sensor continuously scans the whole surface on the air outlet side of the evaporator. The sensed temperature is used to detect any cold spots that may dynamically form on the evaporator surface. The system is controlled in response to the detected temperature.
