The invention discloses a substrate of the synthesis of conjugated linolic acid; the weigh proportion of each components of the culturation base is: 1 share fatty acid linolic acid, 1 share absolute ethyl alcohol, 2 shares glucoses, and 0.65 share soybean powder. The invention also discloses a preparation method of the substrate: 1 portion of fatty acid linolic acid is dissolved in 1 portion of absolute ethyl alcohol, mixed with 2 portions of glucoses and 0.65 portions of soybean powder, and the mixture is thoroughly dissolved by water and sieved by bacteria sieve, which can be used then. The invention further discloses an application method of the conjugated linolic acid synthesized by the culturation base: the substrate is added with high content glucoses, the lactobacillic acid mostly takes glocoses as a carbon source to prevent the lactobacillic acid from being decomposed, and still cells are adopts to synthesize CLA to cut down the carbon sources from being comsumed by cells; LA is dissolved in alcohol and added into the substrate, so as to homogenously distribute LA into the substrate and enhance the catalyzing effect.
