The invention relates to a test method for studying high-
temperature stress on wheat, and belongs to the field of
plant seedling high-
temperature stress tests. The test method comprises the following steps:
seedling culture, variety selection,
sodium hydrosulfide solution preparation,
sodium hydrosulfide solution spraying,
sodium hydrosulfide solution concentration optimization,
sodium hydrosulfide solution spraying time and frequency optimization and the like, wherein
seedling culture comprises seed selection, soaking of seeds in
clean water, seed soaking and sterilization, seed soaking at a constant temperature,
germination acceleration, secondary seed selection, seed arrangement, seedling acceleration, seed picking and seedling.
Seedling selection and optimization is performed multiple times in the period from seed soaking to seedling
transplanting during seedling, therefore, consistency of initial test materials is guaranteed. The
sodium hydrosulfide solutions of different concentration are sprayed to wheat seedlings cultivated for 20 days for high-
temperature stress relieving tests, and concentration, spraying time, spraying frequencies of the
sodium hydrosulfide solutions are optimized; besides, in combination with optimization and innovation of a sampling method during material drawing in the test, the accuracy of final
test data can be better guaranteed from
test design, procedures and results, therefore, great benefit is provided for final disclosure of physiological and biochemical changes after test treatment.
