The invention discloses a method for purifying hydroxytyrosol. The method for purifying the hydroxytyrosol comprises the following steps of: dissolving an oleuropein crude product by using deionized water under stirring; passing through an adsorption type macroporous resin chromatographic column, washing with the deionized water, performing elution by using ethanol aqueous solution, and collecting eluent; performing vacuum concentration until no alcohol exists to obtain refined oleuropein aqueous solution; adding an inorganic acid to react; adding sodium hydroxide to adjust the pH value to be 7 to obtain hydroxytyrosol-containing aqueous solution; performing vacuum concentration; performing extraction by using equivalent ethyl acetate; and laminating ester and concentrating to remove the ethyl acetate to obtain faint yellow paste hydroxytyrosol with purity of more than or equal to 80 percent. In the method, the characteristic of one-way conversion from the oleuropein in an olive leaf resource to the hydroxytyrosol is completely utilized, so that the conversion rate is high, the current situation that the oleuropein component is blindly separated in the purification process of the oleuropein is changed, the oleuropein is converted into the hydroxytyrosol with high economic value, economic benefit is improved, and difficulty in preparation of high-content hydroxytyrosol is greatly reduced.

Preparation method of long-circulation lipidosome of olive polyphenol extract rich in hydroxytyrosol and verbascoside

The invention discloses a preparation method of long-circulation lipidosome of an olive polyphenol extract rich in hydroxytyrosol and verbascoside. Olive leaf is taken as a raw material, and is prepared into an olive polyphenol extract by extracting with 30 to 50 percent of ethanol, separating with a ceramic membrane and an ultrafiltration membrane and concentrating with a nanofiltration membrane; 90 percent of the hydroxytyrosol, 10 to 40 percent of an oleuropein extract and 90 percent of the verbascoside are compounded to prepare the olive polyphenol extract rich in the hydroxytyrosol and the verbascoside; the hydroxytyrosol and verbascoside polyphenol as model materials, lecithin, cholesterol, a Tween-80 surfactant and the like are prepared into the corresponding lipidosome. The lipidosome has the advantages that the bioavailability of the olive polyphenol extract is remarkably increased, and high encapsulation efficiency, uniform particle size distribution, good stability, slow release property, oxidation resistance and an antibacterial effect are achieved. The preparation method is simple and feasible, equipment is simple, and the application of the lipidosome in the medicine field can be widened.

Method and apparatus used for preparing olive leaf extract

The invention discloses a method and an apparatus used for preparing an olive leaf extract. The method comprises the steps that: olive leaves are subjected to ultrasonic vacuum boiling extraction by using a mixed solvent; vacuum pump-filtration is carried out, such that a clarified liquid is obtained; fat-soluble impurities are removed through petroleum ether extraction, and a lower-layer clear solution is obtained; the clear solution is subjected to rotary evaporation until no alcohol smell is left in the solvent; the solution is diluted; the diluant is subjected to macroporous resin purification; the obtained solution is subjected to standing for a period of time, such that oleuropein and flavonoid substances are sufficiently absorbed; the oleuropein is eluted by using an ethanol-water mixed solvent with a proper concentration, and a target liquid is collected; the collected target liquid is subjected to dextrane gel purification; standing is carried out, and an ethanol-water solution is used for eluting; a eluant containing effective components is collected; the collected eluant is subjected to rotary evaporation until no alcohol smell is left; and the obtained material is dried, such that the extract is obtained. The oleuropein content of the extract reaches 85-92%, and the total flavonoid content of the extract reaches 25-35%. The method has the advantages of simple operation, short extraction time, high efficiency, high product recovery rate, good quality, and stable quality.
