DNA amplification and subtraction techniques

A method of isolating genomic or RNA-derived duplex fragments which are unique to one of two fragment mixtures. The fragments in positive-source and negative-source mixtures are separately equipped with end linkers, and each mixture is amplified by successive primed-strand replications, using a single primer which is homologous to the associated linker. The second-source linker is biotinylated, and the fragments in this mixture are hybridized in molar excess with the fragments in the positive-source mixture. DNA species which are not hybridized with the biotinylated species, i.e., species that are unique to the positive-source mixture, are isolated after removal of hybridized species by affinity chromatography. Also disclosed is a method of amplifying a mixture of DNA fragments by repeated linker / primer replication.

Each order is identified and added to a queue for the originating hospital. Orders are reviewed and authorized at remote order processing centers by licensed pharmacy personnel. Computers at the remote order processing centers are linked to hospital pharmacy information systems. A pharmacist at a remote order processing center selects a hospital, reviews orders from the queue for the selected hospital, and enters them directly into the hospital's pharmacy information system. The present invention also supports service level tracking and alerts for aging orders. Commitments are specified in service level agreements. The center then processes the orders according to the commitments. Hospital specific clinical initiatives and policies are stored in a central data repository and made available to personnel to ensure hospital policy compliance.

Shortened purification process for the production of capsular streptococcus pneumoniae polysaccharides

A shortened process for producing a solution containing substantially purified capsular polysaccharides from a cellular Streptococcus pneumoniae lysate broth is described. Ultrafiltering and diafiltering a clarified S. pneumoniae lysate followed by pH adjustment to less than 4.5, preferably about 3.5, precipitated at least 98% of the protein in the solution without seriously affecting polysaccharide yield. Furthermore, following ultrafiltration and diafiltration and acidification to a pH of less than 4.5, filtration using activated carbon precipitated at least 90% of remaining protein without seriously affecting polysaccharide yield. Exemplary, non-limiting S. pneumoniae serotypes that can be purified using the shortened process of the invention are 1, 4, 5, 6A, 6B, 7F, 9V, 14, 18C, 19A, 19F, and 23F. In one embodiment, the Streptococcus pneumoniae cells are lysed using deoxycholate sodium (DOC), while in another embodiment the lytic agent is a non-animal derived lytic agent such as N-lauryl sarcosine sodium (NLS).

In the same display area, a sub-region containing a checklist specific to the one or more images is displayed. Items from the checklist can be de-selected. This checklist comprises a list of selectable items, each item representing an organ, structure, or abnormality that has to be checked by the user. Linked to each item in the checklist is a default statement that is a pre-prepared statement indicative of the normality of the item. The default statement is not displayed by default as part of the checklist. The user can select an item from the checklist for providing comments by dictation thereon responsive to an observation in the radiological image. At the end of the image analysis, an editable itemised medical report is generated containing each item of the checklist as a heading and either the dictated comment or the default statement associated with the item as an observation. The editable itemised medical report is rapidly and accurately generated, information is confined to a single screen reducing the look-away time and general discomfort over time for the user.

In normal practice, a whole blood sample of a patient are provided and then introduced into an apparatus that contains a flow channel. An acoustic field, which contains acoustic standing waves from external ultrasonic transducers, is located within the flow channel. Laminar flow is maintained in the flow channel. Blood cells and platelets are separated from blood plasma or blood serum at the end of the flow channel and collected. The method described herein allows fluid components to differentially migrate to areas of preferred acoustic interaction. The parameters that affect separation of particles are size, density, compressibility of the particles, and the fluid surrounding the particles.

Methods, systems and devices are described for temporarily or permanently evacuating stagnating air from a diseased lung area, typically for the purpose of treating COPD. Evacuation is accomplished by displacing the stagnant CO2-rich air with a readily diffusible gas using a transluminal indwelling catheter specially configured to remain anchored in the targeted area for long term treatment without supervision. Appropriate elevated positive gas pressure in the targeted area is then regulated via the catheter and a pneumatic control unit to force under positive pressure effusion of the diffusible gas out of the area into neighboring areas while inhibiting infusion of other gases thus effecting a gradual gas volume decrease and deflation of the targeted area.
