A ribosome display library of a whole set of single-chain antibody genes against the fenitrothion is constructed by using a ribosome display technology and rounding a hybrid tumor technology, and three single-chain antibody genes with high affinity and high specificity for the fenitrothion are screened from the library. After the single-chain antibody genes are connected with vectors PPOW3.0 and then subjected to soluble expression in escherichia coli, three soluble proteins with molecular weights of about 30KDa are obtained; and by enzyme-linked immuno sorbent assay (ELISA) and Biacore analysis, the three antibody proteins have high affinity and high specificity for the fenitrothion, so excellent single-chain antibody genes and antibody proteins are provided for establishing ELISA and immune sensors. The invention has significance for realizing quick, simple, convenient and accurate detection of organic phosphorus pesticide residue, and provides a new direction.

The method comprises the following steps that (1) the fenitrothion residues in a maize contamination sample are extracted, and residue extracting solutions are obtained; (2) the SERS of all the concentrations of the residue extracting solutions is measured,and thus fenitrothion residue spectrums are obtained; (3) base lines in the fenitrothion residue spectrums are removed through a polynomial fitting algorithm, and the fenitrothion residue spectrums are preprocessed through convolution smoothing and a wavelet transform algorithm; (4) main information of the fenitrothion residue spectrums is extracted through a principal component analysis method, and the detection limit of the SERS to the fenitrothion residues in the maize is judged; and (5) an SVR regression model is established, and the fenitrothion residues in the maize are quantitatively analyzed. According to the method, the SERS and a chemometrics method are combined, and thus the fenitrothion pesticide residues in the maize are detected easily, quickly and accurately.
