A lymphocyte separation medium density gradient centrifugation method is used for separating umbilical cord blood mononuclear cells, a dulbecco modified eagle medium (DMEM)/F12 is used for primary culture of the mesenchymal stem cells of the human umbilical cord blood, and the method increases adherence of the cells, reduces growth of osteoclast like cells remarkably, facilitates formation of human umbilical cord blood-mesenchymal stem cells (hUCB-MSCs) colonies, and greatly improves successful rate of culture of the hUCB-MSCs. The DMEM/F12 is used continuously for subculturing to a P2 generation, the method is combined by a using enzymatic digestion and differential velocity adherent method simultaneously, and the method facilitates purification of P1-P2 generation cells remarkably. A P3 generation and the following generations are cultured by using an Oricell human umbilical cord mesenchymal stem cell culture medium, marker proteins and good morphological characteristics and growth characteristics of the hUCB-MSCs are maintained, and multilineage differentiation potential of the hUCB-MSCs is maintained. In addition, costs of the culture media adopted by the method are reduced remarkably.

The invention discloses a quantum-dot-based method for carrying out in-situ and real-time detection on heavy metal ions in cells. The method comprises the following steps of: A. preparing a carboxymethyl chitosan-CdTe quantum dot solution with certain concentration; B. digesting canine kidney cells overgrowing in a T25 cell culture flask bottle into a unicellular suspension, inoculating into a cell culture dish, and culturing; C. adding the carboxymethyl chitosan-CdTe quantum dot solution into the culture dish, and hatching for a period of time, thereby obtaining the canine kidney cells marked with fluorescence; and D. adding a DMEM (dulbecco's modified eagle medium) nutrient solution containing heavy metal ions with gradient concentration and unknown concentration respectively in washed cells in the culture dish, hatching together with the cells, detecting the average fluorescent intensity of the cells respectively by a flow cytometry, and drawing a standard curve through the changes of the cell fluorescent intensity, so that the concentration of the heavy metal ions in the cells can be calculated. The method is simple and convenient to operate; the fluorescence of marked cells can be quenched regularly as the increase of heavy metal ions, and the real-time and in-situ detection on the outer source heavy metal ions in the cells is realized.

The conditioned medium is obtained by the steps of: culturing BRL-3A cells until the cells are converged, collecting supernatant of the cultured cells, performing centrifugal separation, and filtering by using a filter membrane to obtain the conditioned medium. Compared with the prior art, the method has the advantages that: the long-term subculture of the chicken embryonic stem cells can be realized, the proliferation of the cells is quick, and the cloning yield is high.

According to the preparation method, glede tea, which accounts for 0.6 to 0.8 percent of the total weight of the beverage, is crushed into tea powder, sieved by a 50-mesh sieve and then weighed. After that, water accounting for 8 to 10 times of the weight of the glede tea powders is added into an extraction tank, heated to 75 to 90 DEG C at the room temperature, added with the tea powders for stirring extraction for 8 to 12 minutes. Then the materials are filtered by a filter screen being 200 to 300 meshes to obtain filter liquid (1). Water accounting for 4 to 6 times of the weight of the filter residue is adopted for extracting for 4 to 6 minutes under the same conditions of the filter residue. The filtering operation is carried out by the filter screen being 200 to 300 meshes to obtain filter liquid (2). The filter liquids (1) and (2) are merged and added into the extraction tank. The filter liquid is heated to 60 to 80 DEG C and the pressure thereof is reduced the to -15kPa to 20kPa for concentration. Condensed liquid, i.e. aromatic water, is collected. The collecting of the condensed liquid is stopped until the weight of the condensed liquid is 1 to 1.5 times of that of the tea powder. Then concentration is continuously carryed out until the weight is 1/4 to 1/3 of the total filter liquid weight. The spray drying is carryed out to obtain dry powder. The prepared dry powder, the aromatic water, the white granulated sugar and the citric acid are mixed and prepared. The glede tea beverage has the advantages that the tea beverage has the effects of relieving summer heat and quenching the thirst, helping digestion and enhancing the human body immunity, the quality is easy to control, and in addition, the taste is good.
