Time-Resolved Spectroscopy System and Methods for Multiple-Species Analysis in Fluorescence and Cavity-Ringdown Applications

A time-resolved spectroscopy system employing a time-division multiplexing optical device with no dispersive optical elements to perform lifetime and concentration measurements in multi-species samples, is disclosed. Some examples include fluorescence and cavity ring-down spectroscopy. The system is unique in its compactness and simplicity of operation. In one embodiment, the system makes use of only one photo-detector and an efficient linear regression algorithm. The system offers a measurement time for multiple species measurements of less than 1 s. The system can also be used to perform fluorescence correlation spectroscopy and fluorescence cross-correlation spectroscopy. Four methods to de-convolve a multi-component, exponentially decaying optical signal such as obtained with the system disclosed here, are presented. These methods may be applied to the measurement of fluorescence decay lifetimes and cavity ring-down times, the latter used extensively for the measurement of gas and trace-gas concentrations in complex mixtures, via absorption spectroscopy.

The invention discloses a single-shot multi-information diagnostic target. The diagnostic target is characterized in that a positive plate impacts a substrate at a high speed, and enters a sample through the substrate to conduct shock compression; the shock wave is reflected back and forth between the substrate with high shock impedance and a composite window to compress the sample multiple times so as to generate a high-temperature and high-pressure plasma and send light radiation; the light radiation enters a multi-channel radiation pyrometer and a time-resolved spectrometer through the composite window and a fiber bundle to synchronously measure the shock luminous radiation history and the time-resolved transient spectra; the interface particle velocity of the shock wave changes when the shock wave is reflected between the sample and the window interface; and the particle velocity profile is measured by a Doppler probe system. The state parameters of the sample compressed for multiple times can be determined through correlation analysis of the particle velocity profile and the shock luminous radiation history in combination with the impedance matching method. The diagnostic target has a compact structure, and can achieve the synchronous measurement of multiple state parameters of the plasma in the same single-shot experiment, such as shock wave velocity, particle velocity, pressure, temperature, density and time-resolved transient spectra.

A system, device and / or method for multiplex assays. In a particular, but non-limiting, example there is provided a multiplex array, such as a suspension array. Luminescence decay lifetimes are utilised for probes in a suspension array, and coding / decoding the codes from time-resolved spectra. Lifetime populations can be generated at distinct colour bands. A novel temporal technique or dimension is applied over conventional spectral and intensity combinations, thereby expanding the multiplexing capacity of a suspension array. In one example form, the multiplexing capacity of a suspension array can be expanded to the order of about 58. This provides a reliable, high-throughput and relatively inexpensive solution for multiplex assays in various areas of application such as life sciences, data storage and security.

Provided herein are devices, systems, and methods for characterizing a biological sample in vivo or ex vivo in real-time using time-resolved spectroscopy. A light source generates a light pulse or continuous light wave and excites the biological sample, inducing a responsive fluorescent signal. A demultiplexer splits the signal into spectral bands and a time delay is applied to the spectral bandsso as to capture data with a detector from multiple spectral bands from a single excitation pulse. The biological sample is characterized by analyzing the fluorescence intensity magnitude and / or decayof the spectral bands. The sample may comprise one or more exogenous or endogenous fluorophore. The device may be a two-piece probe with a detachable, disposable distal end. The systems may combine fluorescence spectroscopy with other optical spectroscopy or imaging modalities. The light pulse may be focused at a single focal point or scanned or patterned across an area.

A system for time resolved spectroscopy comprises a CCD device arranged to receive a time varying spectrum. Time resolved spectroscopy is the analysis of a spectrum that varies with time due to any cause. The variation with time of the spectrum may be due, for example, to (i) changes of the spectrum from a given sample space with time (such as due to emission decay); and (ii) changes in the spectrum as successive portions of a sample space are sampled (such as laser scanning microscopy). The CCD device comprises an array of photosites arranged to detect the time varying spectrum and to produce signal charge representative of the spectrum, and one or more CCD multiplication registers arranged to receive the signal charge and to provide charge multiplication. An enhancement of speed and sensitivity is given by a storage area which receives the signal charge in parallel from the array of photosites to provide the charge in serial to the one or more multiplication registers.

A broadband light source coupled to an optical fiber guides light into a gauge via a first leg where a plano-convex lens collimates the light source internally. The invention relates to a super sensitive time resolution spectrograph which comprises an optics unit and an electricity unit. The optics unit comprises an incidence slit, a light collimating part, a concave mirror, a grating light-dividing part, a spatial light modulator and a convergence light-receiving part, the concave mirror comprises a first concave mirror and a second concave mirror, and the electricity unit comprises a random number generator, a single photon point detector, a counter, a time measuring instrument, a control module, a data packet storage and a compression sensing module. To-be-detected quite-weak light incomes through the entrance slit and goes through the light collimating part and the first concave mirror for beam expanding and collimating so as to enable the light to be parallel light, the parallel light is irradiated on the grating light-dividing part, a generated grating reflecting light field is reflected by the second concave mirror to be folded on the spatial light modulator to form a spectrum band, the spatial light modulator randomly modulates the spectrum band to enable emergent light of the spectrum band to be emitted to the convergence light-receiving part at a certain random probability, and the convergence light-receiving part filters stray light and transmits the filtered light to the single photon point detector.

Measuring method and measuring device for time resolution spectrum

The ivnention discloses a new method of new measuring apparatus for measuring time differentiating spectrum which are parallel at time. Light signal travels across monochromator, transformed into electric signal by photomultiplier, attenuation curves with the wavelength can be recorded conveniently by digital storage oscillograph, the data recording attenuation curves with different wavelength is disposed to acquire time differentiating spectrum by assembling luminous intensity of different wavelength at the same attenuation time. Its properties are measuring parallel at time, so that measuring time is shorten greatly and measuring efficiency is increased, besides, oscillograph is fairly usual laboratory apparatus, present apparatus being used fully and decreasing cost.
