Salmonella bacteriophage CKT1 without transduction of drug-resistant gene and application of salmonella bacteriophage CKT1

The invention discloses a salmonella bacteriophage CKT1 without transduction of drug-resistant gene and application of the salmonella bacteriophage CKT1, the virulent bacteriophage CKT1 is preserved in China Center for Type Culture Collection on January 21, 2022, and the preservation number of the virulent bacteriophage CKT1 is CCTCC (China Center for Type Culture Collection) 2022110. The separated salmonella bacteriophage CKT1 has splitting activity on D-group salmonella such as salmonella pullorum, salmonella gallinarum and salmonella enteritidis from different sources, is short in incubation period, high in progeny bacteriophage outbreak amount and low in bacteriophage mutation resistance rate, does not carry virulence genes and drug-resistant genes on a genome, does not have transduction capability, and can be used for preparing the salmonella bacteriophage CKT1. The bacteriophage does not cause transmission of drug-resistant genes, is a safe, efficient and high-specificity virulent bacteriophage, and does not destroy intestinal flora of chicks. The bacteriophage preparation can become a novel environment-friendly auxiliary means for purifying salmonella pullorum in a breeding chicken farm, and is also an alternative strategy for preventing and treating salmonella pullorum or salmonella gallinarum infection of chicken flocks.

Live salmonella vaccine and methods to prevent fowl typhoid

We constructed S. Gallinarum strains deleted for the global regulatory gene fur (FIG. 1) and evaluated their virulence and protective efficacy in Rhode Island Red chicks and Brown Leghorn layers. The fur deletion mutant was a virulent and, when delivered orally to chicks, elicited excellent protection against lethal S. Gallinarum challenge. We also examined the effect of a pmi mutant and a combination of fur deletions with mutations in the pmi and rfaH genes, which affect O-antigen synthesis, and ansB, whose product inhibits host T cell responses. The ΔAfur Δpmi and Δfur ΔansB double mutants were attenuated, but not protective when delivered orally to chicks. However, a Δpmi Δfur strain was substantially immunogenic when administrated intramuscularly. Altogether our results show that the fur gene is essential for virulence of S. Gallinarum and the fur mutant is effective as a live recombinant vaccine against fowl typhoid.

The invention provides a PCR detection kit for rapidly detecting salmonella and identifying salmonella pullorum / salmonella gallinarum and an application thereof. According to the applicant, a primer is designed for a cigR gene, a PCR detection technology is adopted for detecting the cigR gene, and whether a detected object belongs to salmonellae or not or whether the detected object is the salmonella pullorum / salmonella gallinarum or not can be analyzed and judged according to amplification and detection conditions. In a specific scheme, the detection kit at least comprises a forward primer asshown in SEQ ID NO.1 and a reverse primer with a nucleotide sequence as shown in SEQ ID NO.2. The kit is simple, rapid, good in specificity, high in sensitivity and good in repeatability.
