Method for measuring ingestion preference degrees of sea cucumbers

The invention relates to the technical field of animal breeding, in particular to a method for measuring the ingestion preference degrees of sea cucumbers. The invention solves the problem that the food is difficult to observe and count by adding an indicator to natural foods and sediments difficult to distinguish and easy to leach. In the invention, an element analyzer is used to measure the heavy metal contents in the indicator and calculate the ingestion preference degrees of the sea cucumbers so as to reduce errors existing in traditional methods through manual observation and simple weighing and improve the accuracy and the operability of the experiment. The invention considers food availability, is more objective than a method of singly calculating the proportions of various foods in the inclusions of intestinal tracts and has practical application values.

Detection of infectious prion protein by seeded conversion of recombinant prion protein

The present disclosure relates to methods and compositions for the detection of infectious proteins or prions in samples, including the diagnosis of prion related diseases. One embodiment is an ultrasensitive method for detecting PrP-res (PrPSc) that allows the use of recombinant PrP-sen (rPrP-sen) as a substrate for seeded polymerization. A sample is mixed with purified rPrP-sen to make a reaction mix which is incubated to permit aggregation of the rPrP-sen with the PrP-res that may be present in the sample. Any aggregates are intermittently disaggregated by agitation (for example by sonication) and the reaction allowed to proceed to amplify target substrate. Any rPrP-res(Sc) in the reaction mix is detected to indicate the presence of PrP-res in the original sample. This assay, which is called rPrP-PMCA, is surprisingly much faster than existing PMCA methods, yet it still retains sufficient sensitivity to detect extremely low levels of PrP-res. An alternative of rPrP-PMCA is the QUIC method in which shaking of the reaction mixture is substituted for sonication. The surprising speed and efficiency of the method permits the rapid identification and diagnosis of prion disease, which can limit the transmission of prion diseases, particularly through the food supply.

Breeding and fry rearing method of trachidermus fasciatus

The invention relates to the field of trachidermus fasciatus, in particular to a breeding and fry rearing method of trachidermus fasciatus. The method specifically comprises the following steps: parent fish cultivation and wintering period treatment, artificial spawning induction, spawning and fertilized egg collection, egg mass incubation, newly hatched fry cultivation and large-scale fry cultivation and domestication. In the parent fish overwintering, wild living freshwater shrimps need to be caught in advance and cultured in an overwintering pond so as to meet food supply of the trachidermus fasciatus in a whole overwintering period. In the artificial spawning induction, a use method and dosage of an oxytocic are controlled so as to meet requirements of spawning induction of the trachidermus fasciatus. An egg collector used in the spawning is a transparent organic glass product so as to effectively avoid disturbance of a spawning behavior of the trachidermus fasciatus parent fish by turning over the egg collector when the spawning is checked, and can remarkably improve the egg collecting efficiency. The method can effectively improve a parent fish spawning rate, a fertilized egg hatching rate, a fry survival rate and an artificial compound feed domestication success rate, and is complete and efficient for breeding and fry rearing of trachidermus fasciatus.
