Apparatus, method, logic arrangement and storage medium are provided for increasing the sensitivity in the detection of optical coherence tomography and low coherence interferometry (“LCI”) signals by detecting a parallel set of spectral bands, each band being a unique combination of optical frequencies. The LCI broad bandwidth source can be split into N spectral bands. The N spectral bands can be individually detected and processed to provide an increase in the signal-to-noise ratio by a factor of N. Each spectral band may be detected by a separate photo detector and amplified. For each spectral band, the signal can be band p3 filtered around the signal band by analog electronics and digitized, or, alternatively, the signal may be digitized and band pass filtered in software. As a consequence, the shot noise contribution to the signal is likely reduced by a factor equal to the number of spectral bands, while the signal amplitude can remain the same. The reduction of the shot noise increases the dynamic range and sensitivity of the system.

Diffraction grating based interferometric systems and methods

Diffraction grating based fiber optic interferometric systems for use in optical coherence tomography, wherein sample and reference light beams are formed by a first beam splitter and the sample light beam received from a sample and a reference light beam are combined on a second beam splitter. In one embodiment, the first beam splitter is an approximately 50 / 50 beam splitter, and the second beam splitter is a non 50 / 50 beam splitter. More than half of the energy of the sample light beam is directed into the combined beam and less than half of the energy of the reference light beam are directed into the combined beam by the second beam splitter. In another embodiment, the first beam splitter is a non 50 / 50 beam splitter and the second beam splitter is an approximately 50 / 50 beam splitter. An optical circulator is provided to enable the sample light beam to bypass the first beam splitter after interaction with a sample. Two combined beams are formed by the second beam splitter for detection by two respective detectors. More than half of the energy of the light source provided to the first beam splitter is directed into the sample light beam and less than half of the energy is directed into the reference light beam. The energy distribution between the sample and reference light beams can be controlled by selection of the characteristics of the beam splitters.

Camera Adapter Based Optical Imaging Apparatus

The invention describes several embodiments of an adapter which can make use of the devices in any commercially available digital cameras to accomplish different functions, such as a fundus camera, as a microscope or as an en-face optical coherence tomography (OCT) to produce constant depth OCT images or as a Fourier domain (channelled spectrum) optical coherence tomography to produce a reflectivity profile in the depth of an object or cross section OCT images, or depth resolved volumes. The invention admits addition of confocal detection and provides simultaneous measurements or imaging in at least two channels, confocal and OCT, where the confocal channel provides an en-face image simultaneous with the acquisition of OCT cross sections, to guide the acquisition as well as to be used subsequently in the visualisation of OCT images. Different technical solutions are provided for the assembly of one or two digital cameras which together with such adapters lead to modular and portable high resolution imaging systems which can accomplish various functions with a minimum of extra components while adapting the elements in the digital camera. The cost of such adapters is comparable with that of commercial digital cameras, i.e. the total cost of such assemblies of commercially digital cameras and dedicated adapters to accomplish high resolution imaging are at a fraction of the cost of dedicated stand alone instruments. Embodiments and methods are presented to employ colour cameras and their associated optical sources to deliver simultaneous signals using their colour sensor parts to provide spectroscopic information, phase shifting inferometry in one step, depth range extension, polarisation, angular measurements and spectroscopic Fourier domain (channelled spectrum) optical coherence tomography in as many spectral bands simultaneously as the number of colour parts of the photodetector sensor in the digital camera. In conjunction with simultaneous acquistion of a confocal image, at least 4 channels can simultaneously be provided using the three color parts of conventional color cameras to deliver three OCT images in addition to the confocal image.

Fourier domain optical coherence tomography employing a swept multi-wavelength laser and a multi-channel receiver

The present invention is an alternative Fourier domain optical coherence system (FD-OCT) and its associated method. The system comprises a swept multi-wavelength laser, an optical interferometer and a multi-channel receiver. By employing a multi-wavelength laser, the sweeping range for each lasing wavelength is substantially reduced as compared to a pure swept single wavelength laser that needs to cover the same overall spectral range. The overall spectral interferogram is divided over the individual channels of the multi-channel receiver and can be re-constructed through processing of the data from each channel detector. In addition to a substantial increase in the speed of each axial scan, the cost of invented FD-OCT system can also be substantially less than that of a pure swept source OCT or a pure spectral domain OCT system.

An Optical Coherence Tomography (OCT) device irradiates a biological tissue with low coherence light, obtains a high resolution tomogram of the inside of the tissue by low-coherent interference with scattered light from the tissue, and is provided with an optical probe which includes an optical fiber having a flexible and thin insertion part for introducing the low coherent light. When the optical probe is inserted into a blood vessel or a patient's body cavity, the OCT enables the doctor to observe a high resolution tomogram. In a optical probe, generally, a fluctuation of a birefringence occurs depending on a bend of the optical fiber, and this an interference contrast varies depending on the condition of the insertion. The OCT of the present invention is provided with polarization compensation means such as a Faraday rotator on the side of the light emission of the optical probe, so that the OCT can obtain the stabilized interference output regardless of the state of the bend.

Optical coherence tomography with 3d coherence scanning

Optical coherence tomography with 3D coherence scanning is disclosed, using at least three fibers (201, 202, 203) for object illumination and collection of backscattered light. Fiber tips (1, 2, 3) are located in a fiber tip plane (71) normal to the optical axis (72). Light beams emerging from the fibers overlap at an object (122) plane, a subset of intersections of the beams with the plane defining field of view (266) of the optical coherence tomography apparatus. Interference of light emitted and collected by the fibers creates a 3D fringe pattern. The 3D fringe pattern is scanned dynamically over the object by phase shift delays (102, 104) controlled remotely, near ends of the fibers opposite the tips of the fibers, and combined with light modulation. The dynamic fringe pattern is backscattered by the object, transmitted to a light processing system (108) such as a photo detector, and produces an AC signal on the output of the light processing system (108). Phase demodulation of the AC signal at selected frequencies and signal processing produce a measurement of a 3D profile of the object.

Systems and methods for phase measurements

Preferred embodiments of the present invention are directed to systems for phase measurement which address the problem of phase noise using combinations of a number of strategies including, but not limited to, common-path interferometry, phase referencing, active stabilization and differential measurement. Embodiment are directed to optical devices for imaging small biological objects with light. These embodiments can be applied to the fields of, for example, cellular physiology and neuroscience. These preferred embodiments are based on principles of phase measurements and imaging technologies. The scientific motivation for using phase measurements and imaging technologies is derived from, for example, cellular biology at the sub-micron level which can include, without limitation, imaging origins of dysplasia, cellular communication, neuronal transmission and implementation of the genetic code. The structure and dynamics of sub-cellular constituents cannot be currently studied in their native state using the existing methods and technologies including, for example, x-ray and neutron scattering. In contrast, light based techniques with nanometer resolution enable the cellular machinery to be studied in its native state. Thus, preferred embodiments of the present invention include systems based on principles of interferometry and / or phase measurements and are used to study cellular physiology. These systems include principles of low coherence interferometry (LCI) using optical interferometers to measure phase, or light scattering spectroscopy (LSS) wherein interference within the cellular components themselves is used, or in the alternative the principles of LCI and LSS can be combined to result in systems of the present invention.

Simple high efficiency optical coherence domain reflectometer design

The present invention discloses simple and yet highly efficient configurations of optical coherence domain reflectometry systems. The combined use of a polarizing beam splitter with one or two polarization manipulator(s) that rotate the returned light wave polarization to an orthogonal direction, enables one to achieve high optical power delivery efficiency as well as fixed or predetermined output polarization state of the interfering light waves reaching a detector or detector array, which is especially beneficial for spectral domain optical coherence tomography. In addition, the system can be made insensitive to polarization fading resulting from the birefringence change in the sample and reference arms. Dispersion matching can also be easily achieved between the sample and the reference arm for high resolution longitudinal scanning.

A method for accurately synthesizing a full-aperture data map from a series of overlapped sub-aperture data maps. In addition to conventional alignment uncertainties, a generalized compensation framework corrects a variety of errors, including compensators that are independent in each sub-aperture. Another class of compensators (interlocked) include coefficients that are the same across all the sub-apertures. A constrained least-squares optimization routine maximizes data consistency in sub-aperture overlap regions. The stitching algorithm includes constraints representative of the accuracies of the hardware to ensure that the results are within meaningful bounds. The constraints also enable the computation of estimates of uncertainties in the final results. The method therefore automatically calibrates the system, provides a full-aperture surface map, and an estimate of residual uncertainties. Therefore, larger surfaces can be tested with greater departures from a best-fit sphere to greater accuracy than was possible in the prior art.
