PCR diagnostics of dermatophytes and other pathogenic fungi

Dermatophytes which belong to one of the three genera Epidermophyton, Trichophyton and Microsporum are the main cause of fungal infections of skin, hair and nails. Traditional diagnostic procedures consist of microscopy and culture, but due to the slow growth rate of dermatophytes typically two to four weeks are needed before a final diagnosis is obtained. The present invention is a rapid DNA extraction method extracting nucleic acids from fungi (e.g. dermatophytes and other pathogenic fungi) which can be performed from directly on hair, nail or skin specimens from humans, from naturally or experimentally infected animals or from cultured fungal colonies for the use in PCR amplification and detection assays. The present invention also includes specific primer sets for detection of any dermatophyte and for species specific detection of Trichophyton rubrum and Epidermophyton floccosum by PCR and a kit for diagnosing fungal infections.

Nail fungus laser treatment

A method for treating a fungal infection in an infected nail is disclosed. An embodiment includes placing an optical delivery system designed to deliver a laser beam with light having strongly water-absorbed wavelengths to the infected nail. The diseased nail is irradiated with the laser beam, and the fluence and a duration of the laser irradiation received by the nail are adjusted such that by laser energy absorption in a surface portion of the nail a superficial heating of the nail and heat diffusion from said heated surface portion into the infected nail bed are achieved. The bottom of the nail plate in this embodiment is heated to a treatment temperature needed to inactivate the infecting organism.

Topical pyrithione compositions and methods for treatment of nail fungus

Disclosed is a formulation and method for the treatment of fungal infections of the nail (onychomycosis) utilizing a topical composition comprising zinc pyrithione, a solubilizer for the zinc pyrithione, a film former, and a volatile solvent. The formulation is preferably in the form of a lacquer. The composition containing the metal pyrithione complex is topically applied to the infected nail preferably as a nail polish or lacquer which may be applied with a dropper, swab or the like to the afflicted nail surface. The treatment can be done at least two times a day, with a course of treatment lasting from several weeks to several months or until a cure is effected or a significant reduction of the infection is achieved.
