Single-primer nucleic acid amplification methods

The present invention is directed to novel methods of synthesizing multiple copies of a target nucleic acid sequence which are autocatalytic (i.e., able to cycle automatically without the need to modify reaction conditions such as temperature, pH, or ionic strength and using the product of one cycle in the next one). In particular, the present invention discloses a method of nucleic acid amplification which is robust and efficient, while reducing the appearance of side-products. The method uses only one primer, the “priming oligonucleotide,” a promoter oligonucleotide modified to prevent polymerase extension from its 3′-terminus and, optionally, a means for terminating a primer extension reaction, to amplify RNA or DNA molecules in vitro, while reducing or substantially eliminating the formation of side-products. The method of the present invention minimizes or substantially eliminates the emergence of side-products, thus providing a high level of specificity. Furthermore, the appearance of side-products can complicate the analysis of the amplification reaction by various molecular detection techniques. The present invention minimizes or substantially eliminates this problem, thus providing an enhanced level of sensitivity.

System and method for intrusion prevention in a communications network

A method, system and program for preventing intrusion in a communications network. A source node initiates a request for network services, such as session establishment, database access, or application access. Known network resources and authorized user information is stored in a database at a network portal along with access policy rules that are device and user dependent. Identification of the source node is required before the source node can construct a transformed packet header that is included with a synchronization packet before transmission to a destination node. An appliance or firewall in the communications network receives and authenticates the synchronization packet before releasing the packet to its, intended destination. The authentication process includes verification of the access policy associated with the source node. Once received at the destination node, the transformed packet header is reformed by extracting a key index value. The extracted key index is subsequently used to transform the packet header in the response transmitted to the source node.

A gas fueled lighter having a compact storage configuration so as to be easily carried in a pocket or purse, the lighter actuated by sliding a longitudinal portion engaged to the body so as to extend said portion to form an extension having a free end, the free end configured to initiate a flame upon full extension from the body, the flame fueled by gas in the body and ignited by a piezo ignition switch. The preferred embodiment of the present invention is designed to provide an inexpensive, compact, easy to implement lighter providing the flame from an extension when in use, the extension sliding back to form a portion of the body when not in use. This safety feature not only provides a flame extended from the user's hand and body, it also provides an extension for lighting pilot lights, gas grills, fireplaces, and the like. The body of the preferred embodiment has a gas reservoir therein as well as the ignition and fuel valve, and further has a generally rectilinear configuration with at least a portion of the top configured to slide longitudinally to form the extension, the upper face of the top including a frictional area for placement of the user's thumb thereupon for sliding of the extension and initiation / cessation of the flame. The body may be rubberized for a better grip, and may include finger ridges for an enhanced grip of the system, and the overall body ideally has an ergometric design to facilitate comfortable and tactile operation.

Process for upgrading hydrocarbon feedstocks using solid adsorbent and membrane separation of treated product stream

A process for upgrading crude oil fractions or other hydrocarbon oil feedstreams boiling in the range of 36° to 520° C., and preferably naphtha and gas oil fractions boiling in the range of 36° to 400° C., employs a solid adsorption material to lower sulfur and nitrogen content by contacting the hydrocarbon oil, and optionally a viscosity-reducing solvent, with one or more solid adsorbents such as silica gel or silica, silica alumina, alumina, attapulgus clay and activated carbon in a mixing vessel for a predetermined period of time; passing the resulting slurry to a membrane separation zone, optionally preceded by a primary filtration step (i.e., single stage or multiple stages), to separate the solid adsorption material with the adsorbed sulfur and nitrogen compounds from the treated oil; recovering the upgraded hydrocarbon product having a significantly reduced nitrogen and sulfur content as the membrane permeate; mixing the solid adsorbent material with one or a combination of aromatic solvents such as toluene, benzene, the xylenes and tetrahydrofuran to remove and stabilize the sulfur and nitrogen compounds; transferring the solvent to a fractionation tower to recover the solvent, which can be recycled for use in the process; and recovering the hydrocarbons that are rich in sulfur and nitrogen for processing in a relatively small high-pressure hydrotreating unit or transferring them to a fuel oil pool for blending.
