The invention discloses a device for a cell membrane permeabilization migration test and a forming mould for manufacturing the device, belonging to the devices for cell experiments. The device is characterized by comprising a disc-shaped culture dish filled with gelatinized agarose, a cell hole defined by the gelatinized agarose in the culture dish, a chemotaxin hole defined by the gelatinized agarose in the culture dish, a migration channel defined by the gelatinized agarose in the culture dish and communicating the cell hole and the chemotaxin hole, and a macromolecule filter membrane blocked in the migration channel. The forming mould for manufacturing the device is characterized by comprises a base plate, and a mould body, wherein two sides of the base plate are provided with arc-shaped vertical edges, the convex-column-shaped mould body is arranged in the center of the base plate, and manufactured according to the shapes and sizes as well as the structures of the cell hole, the chemotaxin hole, the migration channel and the macromolecule filter membrane. Through trial, compared with the prior art, the device and the mould have the advantages that the migration movement of the cells passing through the porous polyester fiber membrane can be directly observed, the number of the cells successfully passing through the porous polyester fiber membrane can be accurately counted, and the cells successfully passing through the porous polyester fiber membrane can be conveniently collected for follow-up experiments.
