The invention relates to an extracting and purifying method of a chlorella lutein protein complex. The extracting and purifying method comprises the following steps: carrying out wall-breaking treatment onto chlorella by utilizing an ultrasonic crushing method; carrying out solubilization onto chromoprotein on a thylakoid membrane by adopting a surfactant; and purifying the lutein protein complex by adopting ammonium sulfate classification and sedimentation, ion exchange chromatography and gel filtration chromatography. A pigment composition of the complex is analyzed by utilizing full-wavelength scanning and high performance liquid chromatography, wherein the obtained lutein protein complex has remarkable ABTS radical-scavenging activity. The extracting and purifying method disclosed by the invention can ensure structure and function stability of the complex by utilizing surfactant solubilizing chromoprotein; purified lutein protein complex obtained by column chromatography is simple in purifying process and convenient to produce and expand; and the lutein existing in the form of the complex not only improves stability, but also keeps antioxidant activity.

The invention provides a chlorella culture method capable of increasing lutein content and an apparatus. The method comprises (1) primary heterotrophic step, namely adding a heterotrophic culture solution having the pH of 6-8 to a fermentation tank, inoculating the fermentation tank with the chlorella, and culturing until the chlorella cell density is within the range of (5-50)*10<6> / mL, wherein during culturing, the temperature is 27-32 DEG C, the pH is 6-8, and the DO value is 20%-40%; (2) autotrophic-heterotrophic stage, namely, transferring the primary heterotrophic culture solution obtained in the step (1) to autotrophic culture, wherein the culture temperature is 27-32 DEG C, ventilatory capacity of carbon dioxide is 0.03-0.05VVM, and the DO value is controlled within the range of 20%-40%, when the increase amplitude of the lutein content is lower than 1.5-3% / h, transferring the culture solution obtained after autotrophic culture into a fermentation tank for heterotrophic culture for 18-36h, and then returning to the autotrophic culture stage, and the autotrophic-heterotrophic loop is realized in the way.

The invention provides a method used for extracting and separating xanthophyll in marigold. The method comprises following steps: firstly, enzyme is adopted for hydrolysis of marigold flower powder soas to obtain an enzymatic hydrolysate; the enzymatic hydrolysate is subjected to salting out extraction; an obtained product is allowed to stand or is subjected to centrifugation phase separation soas to obtain an upper organic solvent phase and a lower inorganic salt water phase, and the upper organic solvent phase is subjected to evaporation so as to obtain xanthophylls. Compared with conventional extraction method, the advantages are that: the advantages of enzyme-salting out extraction and enzyme-microwave auxiliary salting out extraction are adopted, direct extraction of xanthophyll infresh marigold flowers is realized, steps of fresh marigold flower fermentation, granulation pretreatment process are avoided; and in addition, salting out extraction is capable of realizing phase separation, so that extraction and primary extraction on xanthophyll are realized, primary purifying of target products is realized, enzyme and microwave action is capable of increasing effective component extraction yield effectively, simplifying production technology, and shortening production period.
