Methods for analyzing inter-cellular phenomena

Aspects of the present invention encompass the collection of multispectral images from a population of objects, and the analysis of the collected images to measure at least one characteristic of the population, using photometric and / or morphometric features identifiable in the collection of images. In an exemplary application, the objects are biological cells. In a particularly preferred, but not limiting implementation, the plurality of images for each individual object are collected simultaneously. In an empirical study, the characteristic being measured involves the synapse between conjugated cells. The conjugated cells may represent a subpopulation of the overall population of objects that were imaged. In a particularly preferred, yet not limiting embodiment, the present invention enables the quantization of the redistribution of cellular molecules due to the conjugation of different biological cells. Significantly, such quantization is not feasible with standard microscopy and flow cytometry.

Grating-based sensor combining label-free binding detection and fluorescence amplification and readout system for sensor

A grating-based sensor is disclosed that has a grating structure constructed and designed for both evanescent resonance (ER) fluorescence detection and label-free detection applications. Some embodiments are disclosed which are optimized for ER detection in an air mode, in which the sample is dry. Other embodiments are optimized for ER detection in liquid mode, in which the sample is suspended in liquid medium such as water. One and two-dimensional gratings are also disclosed, including gratings characterized by unit cells with central posts, central holes, and two-level, two-dimensional gratings. A readout system for such sensors is also disclosed. One embodiment includes a first light source optimized for collecting label-free detection data, a second light source optimized for collecting ER fluorescence amplification data, and at least one detector. In one embodiment, the detector is an imaging system and includes a CCD camera for collecting both ER and label-free data. In other embodiments, the at least one detector takes the form of a spectrometer for collection of label-free data and a photomultiplier for collecting ER data. In other embodiments, a single light source such as a tunable laser or broad band light source is used.

The invention provides materials and methods for modulating adenoviral tropism for hepatocytes and other cell types such as splenocytes. It relates to the findings that hypervariable regions (HVRs) of the viral hexon protein interact with the Gla domain of the blood clotting factor FX as part of the infective process in vivo. The invention provides means to disrupt the interaction between hexon and FX, thus reducing infection of hepatocytes and splenocytes, as well as use of targeting agents comprising the Gla domain or a fragment thereof to direct adenoviral vectors to desired target cell or tissue types.

The invention relates to an improved method for predicting the onset of, diagnosing, prognosticating and/or treating dementing diseases. The method comprises determining the level of heme oxygenase-1 suppressor (HOS) activity and/or factor in tissue or body fluid obtained from a patient, and comparing said level with the corresponding level of HOS activity and/or factor in corresponding tissue or body fluid obtained from at least one control person. The tissue or body fluid is suitably blood, plasma, lymphocytes, cerebrospinal fluid, urine, saliva, epithelia or fibroblasts. The method is useful where the dementing disease is any of Alzheimer Disease, Age-Associated Cognitive Decline, Mild Cognitive Impairment, Parkinson disease with dementia, Progressive Supranuclear Palsy, Vascular (i.e. multi-infarct) Dementia, Lewy Body Dementia, Huntington's Disease, Down's syndrome, normal pressure hydrocephalus, corticobasal ganglionic degeneration, multisystem atrophy, head trauma, neurosyphilis, Creutzfeld-Jacob disease and other prion diseases, HIV and other encephalitides, and metabolic disorders such as hypothyroidism and vitamin B12 deficiency. The method may also prove useful in differentiating the“pseudodementia” of depression from Alzheimer disease.
