The invention provides an
inoculation method for identifying the
disease resistance to sugarcane
mosaic virus. The method mainly comprises the following steps: inoculating
virus source living bodies which propagate are stored on a sugarcane
plant; before the
inoculation, young
disease leaves of the
plant are
cut into fine pieces, ground into a homogenate in the presence of a
phosphate buffer solution and filtered with double-layer gauze to obtain a filtrate which is the
virus inoculant; in a cool
evening during the sugarcane growth period, the aerial part of the sugarcane
plant are rapidly
cut along the
soil surface with a sharp blade or scissors, a drop of
virus inoculant is sucked immediately with a dropper and dropped into the underground
wound surface of the sugarcane so that the virus inoculant contacts with the
wound surface and viruses invades the plant; and the
wound surface is uncovered after being shaded for 24 hours. With the method, sugarcane
mosaic virus after being inoculated develops remarkably and uniformly, the sensitivity is high, and the result stability is good. The method has the advantages that the process is simple, rapid, standardized and practical; the manual
rubbing of sugarcane leaves is obviated, and the head scuffing is prevented so that the
operability is higher and the
inoculation efficiency is higher; and the identification result is closer to the field naturally-infected result so that the method is suitable for rapidly carrying out the identifying and the screening of
mosaic virus resistance to large-scale material.
