Novel and efficient syntheses create novel piperazinone intermediates which facilitate the production and use of PNAs. Such syntheses and the products enhance the feasibility of a system which permits the rapid identification of PNA oligomers useful as therapeutics, diagnostics and / or gene characterization tools. A first component of the system is a universal PNA library that most preferably incorporates one or more universal nucleotide bases into carefully selected positions within each oligomer species thereby providing the library with the screening ability of a much larger library. The second component of the system is a high throughput screening system that includes a number of assays designed to provide information on the binding activities of the different PNAs to a target nucleotide sequence (generally, a DNA or RNA sequence). The third component is a software system especially designed to provide rapid analysis of the data collected from the high throughput screening system and to determine therefrom the sequence base identities and sequence lengths of PNA oligomers most likely to bind to and appropriately affect the target molecule.

The present invention provides a linker preferably used when constructing an mRNA / cDNA-puromycin-protein conjugate used in an in vitro virus method, and an mRNA / cDNA-puromycin-protein conjugate constructed using that linker. More specifically, the present invention provides a linker for ligating mRNA and puromycin or a puromycin-like compound to construct an mRNA / cDNA-puromycin-protein conjugate, the linker comprising a single-stranded RNA as a main backbone, and having, in this main backbone, a solid phase binding site for binding an mRNA-puromycin-protein conjugate to a solid phase site, and a pair of cleavage sites provided at locations surrounding the solid phase binding site; an mRNA-puromycin-protein conjugate constructed using this linker; an mRNA bead or an mRNA chip comprising this conjugate; a protein chip produced from this mRNA chip; and a diagnostic kit using the mRNA bead or the mRNA chip.

The invention relates to methods of increasing the titre of a protein of interest in a cell as well as the improved production and purification of optimised biomolecules, one component of which is the domain CH3. A frequently observed effect in biomolecules is the cleaving of the C-terminal amino acid(s), e.g. the C-terminal lysine. The usually incomplete processing of the heavy chain of antibodies for example leads to product heterogeneity. To prevent this product heterogeneity the corresponding codon of the C-terminal lysine of the heavy antibody chain has been deleted by recombinant DNA technology. These optimised antibodies lead to a product titre which is higher than in the wild-type. In addition, they prove advantageous during purification by having better elution characteristics as a result of the reduced charge heterogeneity.
