Method for preparing cell cultures from biological specimens for chemotherapeutic and other assays

An improved system for screening a multiple of candidate therapeutic or chemotherapeutic agents for efficacy as to a specific patient, in which a tissue sample from the patient is harvested, cultured and separately exposed to a plurality of treatments and/or therapeutic agents for the purpose of objectively identifying the best treatment or agent for the particular patient. Specific method innovations such as tissue sample preparation techniques render this method practically as well as theoretically useful. One particularly important tissue sample preparation technique is the initial preparation of cohesive multicellular particulates of the tissue sample, rather than enzymatically dissociated cell suspensions or preparations, for initial tissue culture monolayer preparation. With respect to the culturing of malignant cells, for example, it is believed (without any intention of being bound by the theory) that by maintaining the malignant cells within a multicellular particulate of the originating tissue, growth of the malignant cells themselves is facilitated versus the overgrowth of fibroblasts or other cells which tends to occur when suspended tumor cells are grown in culture. Practical monolayers of cells may thus be formed to enable meaningful screening of a plurality of treatments and/or agents. Growth of cells is monitored to ascertain the time to initiate the assay and to determine the growth rate of the cultured cells; sequence and timing of drug addition is also monitored and optimized. By subjecting uniform samples of cells to a wide variety of active agents (and concentrations thereof), the most promising agent and concentration for treatment of a particular patient can be determined. For assays concerning cancer treatment, a two-stage evaluation is contemplated in which both acute cytotoxic and longer term inhibitory effect of a given anti-cancer agent are investigated.

Intelligent distribution network reconstruction method concerning distributed power generation and energy storage unit

The invention discloses an intelligent distribution network reconstruction method concerning a distributed power generation and energy storage unit. The method comprises the following steps: building a multi-target multi-constraint mathematical model for distribution network reconstruction, encoding control variables in the model, then adopting the improved NSGA-II (Nondominated Sorting Genetic Algorithm II) for solution, repeatedly calling a three-phase load flow calculation module in the calculation process, decoding individuals of all generations of population in the NSGA-II, conducting three-phase load flow calculation to the decoded network reconstruction schemes to further obtain the adaptation degree of all the network reconstruction schemes, and feeding back to the NSGA-II for optimal treatment. The multi-target multi-constraint mathematical model built in the invention can simultaneously concern the influences of the distributed power supply and energy storage unit; the adopted multi-supply three-phase forward-backward sweep method can well put the three-phase unbalance characteristic of the distribution network into consideration, and is high in computing speed; the quick NSGA with the out-of-limit rejection strategy can avoid weight setting blindness, can promptly eliminate infeasible solution and is high in computational efficiency.

Method of optimizing the design, stimulation and evaluation of matrix treatment in a reservoir

A method for designing acid treatments provides for the selection of optimal treatment for well stimulation wherein reservoir characteristics are obtained to further select the reaction kinetic data on the minerals of interests, the treatment to the reservoir is scaled up using a mathematical model and real time damage are computed based on bottomhole pressure and injection rate and compared to that predicted by the mathematical model to adjust the treatment. The model generated facilitates optimization of matrix treatments by providing a rapid quantitative evaluation of various treatment strategies for a formation. Stimulation with non-traditional fluid recipes containing mixtures of inorganic and organic acids, and chelating agents can be readily computed. The computed values can then be used in an economic model to justify the additional costs associated with the use of the non-traditional fluids. Apart from optimizing matrix treatments, the method can be used as a development tool for new fluid systems, as a tool for prediction and removal of inorganic scale and for fluid compatibility testing such as that required in water flooding projects.
