Method and apparatus for imaging a sample on a device

InactiveUS6252236B1Good depth discriminationChemical library matterPhotometrySize determinationResponse radiation
A method and apparatus for imaging a sample are provided. An electromagnetic radiation source generates excitation radiation which is sized by excitation optics to a line. The line is directed at a sample resting on a support and excites a plurality of regions on the sample. Collection optics collect response radiation reflected from the sample I and image the reflected radiation. A detector senses the reflected radiation and is positioned to permit discrimination between radiation reflected from a certain focal plane in the sample and certain other planes within the sample.

Bead dispensing system

A bead dispensing system is provided for delivering small amounts of substances onto substrates. The system can include, for example, a movable support structure having an array of spaced-apart projections depending from its lower side. An attraction source, such as a vacuum, magnetic, and / or electrostatic force, is operable at each projection end region to attract and retain one bead. The projection array can be aligned with an array of bead-receiving regions of a substrate, e.g., an array of spaced-apart wells of a micro-plate or card. In one embodiment, a plurality of reagent-carrying beads are picked up, retained at respective projection end regions, and moved to a location over a multi-well plate. The beads are then released in a fashion permitting each bead to land in a respective well. The system of the invention is particularly useful for fabricating arrays of reagents.

Microfluidic device

A novel approach for fabricating Monolithic Internal micro Pillars (MIPi) made of SU-8 photoresist is described. A microfluidic chip with the internal pillars (a MIPi chip) was used for cell capturing study. The surface of MIPi was coated with specific antibody and then used for capturing cells by affinity binding. An antibody, anti-EGFR, which has high affinity to lung cancer cells, CL1-5, was coated on the micro pillars. The coated MIPi chip specifically captured the cancer cells that were pumped through the MIPi chip. Simulation and experiment was carried out to compare the effect of different geometry of the micro pillars on the cell capturing rate.

Synthesis and use of organosilica particles

Highly functionalized, porous organosilica particles and methods of their synthesis are described that employ high amounts of functional silane such as 3-mercaptopropyl trimethoxysilane. Silane particle diameters are controlled from less than 1 μm to over 100 μm. The particles have a high surface area due to their advantageous internal structures, which consist of large pores, typically up to 10 μm that are linked by small channels of typically about 20 nm diameter. Isothiocyanate modified fluorescent dyes can enter and react with thiol groups inside the pores. The invention also provides clear silica shells of controlled thicknesses to protect fluorescent signals and provide an independent parameter for distinguishing particle types based on light scattering off different sized particles. The particles are stable and useful for many purposes, particularly for optical bar coding in combinatorial synthesis of polymers such as nucleic acid, polypeptide, and other synthesized molecules.

High-throughput mass-spectrometric characterization of samples

The invention relates to the characterization of samples which are located in their many hundreds up to tens or hundreds of thousands on a sample support plate in a regular pattern, a so-called array, by ionization with matrix-assisted laser desorption and mass spectrometric measurement, for example.
