The method is applicable to Brassica napus rape varieties in rape production areas in the Yangtze river basin. By means of the method, the oil content is increased and lodging resistance is improved simultaneously under the condition that the yield of the Brassica napus rapes is higher through high-nitrogen culture, so that quality of the rapes is improved easily, application and popularization of the mechanical harvesting technology are facilitated, and economic benefits of rape production are improved.

Tissue culture and rapid propagation method of new variety 'Shirley' of vertical branch type chionanthus retusus

The invention discloses a tissue culture rapid propagation method of a new variety 'Shirley' of a vertical branch type chionanthus retusus. The method comprises the following steps of (1) cutting a tender stem with the length of 2-2.5 cm from a 'Shirley' stock plant as a test material, inoculating the test material into a culture medium for inducing stem differentiation, and inducing adventitiousbud differentiation and proliferation; (2) when the test material in the step (1) has 4-5 new leaves and is differentiated to form 1-2 adventitious buds, transferring the test material into the culture medium for inducing stem elongation so as to promote stem elongation; (3) when the stem of the test material in the step (2) elongates to 3-5cm, transferring the stem into a rooting culture medium,and carrying out adventitious root induction; and (4) hardening and transplanting rooted seedlings. When the method disclosed by the invention is used for propagating the 'Shirley', the new variety 'Shirley' can be popularized in a short time, so that the new variety 'Shirley' can be rapidly applied to greening practice. A theoretical basis is provided for establishing a rapid propagation system with stable and efficient chionanthus retusus characters and a subsequent genetic transformation experiment.

Application of the broccoli wound-inducible promoter of glucose inhibition of root elongation 1 gene in transgenic plants

In this study, we used a wound-inducible promoter of the broccoli (Brassica oleracea var. italica) GLUCOSE INHIBITION of ROOT ELONGATION1 (GIR1) gene fused to β-glucuronidase (GUS, pBoGIR1::GUS) as a selectable marker. Transgenic broccoli plants expressing pBoGIR1::GUS appear blue in planta at wounded regions after GUS staining for 30 min. Similarly, the blue color is visible in transgenic Arabidopsis and rice plants expressing pBoGIR1::GUS at wounded areas after GUS staining for 2 h, indicating that this promoter is wound-inducible in both dicots and monocots. GUS staining is very rapid and the partial wounding in this study is a nondestructive method that does not affect further plant growth and development. Thus, pBoGIR1::GUS could serve as an effective substitute for antibiotic- and herbicide-resistance genes in the generation of genetically modified crops.
