This invention provides methods and compositions useful for identifying and diagnosing rare fetal cells in a mixed cell population such as a maternal blood sample. The methods entail the use of specific nucleic acid probes that hybridize to fetal cell associated RNAs to identify the rare fetal cells or antibodies that bind to polypeptides encoded by the fetal cell associated RNAs for fetal cell detection. The cells detected by the methods of the present invention are useful for diagnosing the fetal cells for a genetic trait of interest, such as trisomy 21. Novel methods for simultaneous screening for fetal cells and diagnosing the fetal cells are also provided. Compositions comprising the fetal cell associated nucleic acids of the invention and their encoded proteins are also provided. The present invention further provides kits useful for practicing the present methods.

Stem cell mediated treg activation/expansion for therapeutic immune modulation

Disclosed are cells, methods of modulating cells, and therapeutic uses of the cells for the immune modulation of mammals in need thereof. Immune modulation including alteration of cytokine profile, cytotoxic activity, antibody production and inflammatory states is achieved through the administration of various cell types that have been unmanipulated or manipulated in order to endow specific biological activity. Cellular subsets and administration of the subsets in combination with various agents are also provided. One embodiment teaches the previously unknown finding that adipose tissue derived mononuclear cells contain T cells with immune regulatory properties that alone or synergistically with various stem cells induce immune modulation upon administration. Another embodiment is the finding that stimulation of stem cell activation results in stem cell secondary activation of immune modulatory cells, one type which is T regulatory cells (Tregs). One specific embodiment involves extraction of a heterogenous stem cell pool, which contains T regulatory cells, treatment in culture of the population with agents known to stimulate stem cell activation, then subsequent extraction and administration of the purified Tregs. Other embodiments include expansion of Tregs in the presence of antigen in order to generate anti-specific Tregs.

Described here is an automated robotic device that isolates circulating tumor cells (CTCs) or other biological structures with extremely high purity. The device uses powerful magnetic rods covered in removable plastic sleeves. These rods sweep through blood samples, capturing, e.g., cancer cells labeled with antibodies linked to magnetically responsive particles such as superparamagnetic beads. Upon completion of the capturing protocol, the magnetic rods undergo several rounds of washing, thereby removing all contaminating blood cells. The captured target cells are released into a final capture solution by removing the magnetic rods from the sleeves. Additionally, cells captured by this device show no reduced viability when cultured after capture. Cells are captured in a state suitable for genetic analysis. Also disclosed are methods for single cell analysis. Being robotic allows the device to be operated with high throughput.
