Multiple-wavelength spectroscopic quantitation of light-absorbing species in scattering media

An oxygen concentration measurement system for blood hemoglobin comprises a multiple-wavelength low-coherence optical light source that is coupled by single mode fibers through a splitter and combiner and focused on both a target tissue sample and a reference mirror. Reflections from both the reference mirror and from the depths of the target tissue sample are carried back and mixed to produce interference fringes in the splitter and combiner. The reference mirror is set such that the distance traversed in the reference path is the same as the distance traversed into and back from the target tissue sample at some depth in the sample that will provide light attenuation information that is dependent on the oxygen in blood hemoglobin in the target tissue sample. Two wavelengths of light are used to obtain concentrations. The method can be used to measure total hemoglobin concentration [Hb.sub.deoxy +Hb.sub.oxy ] or total blood volume in tissue and in conjunction with oxygen saturation measurements from pulse oximetry can be used to absolutely quantify oxyhemoglobin [HbO.sub.2 ] in tissue. The apparatus and method provide a general means for absolute quantitation of an absorber dispersed in a highly scattering medium.

The reaction cassette for the glycated hemoglobin meter includes: a first zone receiving a first reagent and a blood sample; a second zone receiving a second reagent; a reaction zone in which the blood sample reacts with the first reagent, or through which the second reagent passes to react with a first blood sample mixture obtained by reacting the blood sample with the first reagent; and a measurement zone measuring an amount of total hemoglobin in the first blood sample mixture, or measuring an amount of glycated hemoglobin in a second blood sample mixture obtained by reacting the first blood sample mixture with the second reagent, wherein the blood sample, the first reagent, and the second reagent move between the reaction zone and the measurement zone according to a rotation angle of the reaction cassette when the reaction cassette is rotated. Therefore, since the reaction cassette reduces the operating time, it is possible to resolve storage and distribution problems of the reaction cassette, which occur when reagents are stored in the reaction cassette.

Bone oxygenation measurement

A bone oximeter probe includes an elongated member and a sensor head at an end of the elongated member to make measurements for a bone. The measurements can indicate the viability or nonviability of the bone. In an implementation, the probe is advanced through an incision in soft tissue, towards the underlying bone, and positioned so that the sensor head faces the bone to be measured. Optical signals are sent from the sensor head and into the bone. The bone reflects some of the optical signals which are then detected so that measurements for the bone can be made. Some of these measurements include an oxygen saturation level value, and a total hemoglobin concentration value of the bone.

Device and method for measuring hemoglobin

The present invention relates to a system for measuring the hemoglobin concentration in whole blood, wherein the system comprises: a light-radiating unit including a light source that emits two types of incident light having different wavelengths; a diffusion unit which diffuses the incident light emitted by the light-radiating unit; a cuvette-holding unit which is formed so as to hold a cuvette including a blood sample; a detection unit which detects each absorbance of the two types of incident light having different wavelengths; a processing unit which determines the hemoglobin concentration in the blood by processing the measured absorbance result; and a control unit which regulates the two types of incident light having different wavelengths in order to repeatedly/sequentially radiate same. Although the system for measuring hemoglobin in whole blood of the present invention uses a small amount of whole blood, it is possible to measure the total hemoglobin concentration in an accurate and reliable manner. The system of the present invention aligns the paths of two types of incident light having different wavelengths passing through a microcuvette by using a diffuser plate so as to easily align a light source and increase the reliability of the results. Also, the system of the present invention uses two wavelengths so as to rapidly and accurately measure the total amount of hemoglobin, including oxidized and reduced hemoglobin.

Methods for assaying percentage of glycated hemoglobin

The invention provides enzymatic methods for direct determination of percentage of glycated hemoglobin in blood samples without the need of a separated measurement of total hemoglobin content in blood samples. The methods utilizes one or two different types of oxidizing agents which selectively oxidize low-molecular weight reducing substances and high-molecular weight (mainly hemoglobin) reducing substances in blood samples, coupled with enzymatic reactions catalyzed by proteases, fructosyl amino acid oxidase. The amount of hydrogen peroxide generated in the reaction is measured for determination of percentage of glycated hemoglobin in blood samples. The invention provides kits for performing the methods of the invention.
