[0116] 3.1 In vitro cytocompatibility test

[0117] It has been reported in previous literature that cell adhesion is the basis for cell function and tissue integration in vivo, and cell proliferation is closely related to the amount of new bone formation; good cell adhesion and proliferation can promote a large amount of bone formation around materials (such as orthopedic / dental implants). Production of bone tissue. Therefore, a common method for evaluating the cytocompatibility of materials in vitro is to detect the ability of cells to adhere and proliferate on the surface of the material. In this study, we verified the adhesion and proliferation of hBMSCs on the surface of Dex / Mino liposome-modified PEEK, which is the key to judging whether the material can be applied in the biomedical field.

Abstract

The invention discloses a lipidosome-based method for modifying a PEEK (polyetheretherketone) surface with hexadecadrol / minocycline and an application of the method. Lipidosome loaded with hexadecadrol and minocycline is prepared with a thin film dispersion method and then modifies the PEEK surface. The method comprises following steps: (1), hexadecadrol and lipids are dissolved in a methanol and chloroform mixture (1:1, v / v); (2), methanol and chloroform are removed by a rotary vacuum evaporator, and then a lipid membrane is hydrated by a hydration solution; lipidosome loaded with hexadecadrol is obtained by water bath ultrasonic treatment, and is extruded by a porous polycarbonate membrane; dialysis is performed overnight; (3), lipidosome is mixed with a minocycline storage solution, the mixture is shaken, and lipidosome loaded with hexadecadrol / minocycline is obtained, and is stored for standby application at 4 DEG C after being dialyzed overnight; (4), PEEK implant is put in a dopamine solution for reaction, and ultrasonic cleaning is performed; (5), the PEEK implant is added to a hexadecadrol / minocycline lipidosome solution for soaking and is slightly cleaned, and a product is obtained. The obtained product can promote bone repair, regeneration and integration.

Description

technical field [0001] The invention relates to a liposome-based active small molecule surface modification method and its application, belonging to the technical field of drug controlled release and biomaterials, in particular to liposome-based modification of dexamethasone / minocycline on the surface of PEEK method and application. Background technique [0002] At present, bone defects caused by trauma, bone tumors, infection, inflammation, etc. are still a rapidly growing serious challenge in the medical field worldwide; clinically, orthopedic / dental implants are commonly used hard tissue substitutes for bone defect repair . [0003] PEEK is the material of choice to replace traditional metal implants made of titanium and its alloys. It is a semi-crystalline thermoplastic polymer material with good mechanical properties, chemical stability and thermal stability. Since it was approved by the US Food and Drug Administration in the late 1990s, PEEK has been widely used as a...
