Embodiment 1

[0025] The preparation of embodiment 1 transfer factor

[0026] (1) The method of this application:

[0027] 1. Carefully remove the fat and fascia of the fresh or thawed spleen of healthy laying hens, wash it with normal saline, drain it, cut it into pieces, and weigh it for later use.

[0028] 2 Take the treated spleen and add an equal amount of normal saline according to its weight, and grind it continuously with a colloid mill for 2-3 times, each time for 3 minutes.

[0029] 3 Adjust the pH to 3.5 with 1mol / L HCl at 20°C.

[0030] 4 Put the homogenate with a pH of 3.5 at -20°C and completely freeze it, then completely thaw it with tap water at about 25°C, and repeat freezing and thawing 3 times.

[0031] Centrifuge at 5 6000 rpm for 20 minutes at 4°C, and take the supernatant for later use. Take the residue and add the same amount of normal saline as the spleen of the initial laying hen, grind it with a colloid mill for 3 minutes, adjust the pH to 3.5 again, repeat free...

Embodiment 2

[0038] Embodiment 2 is to the detection of the transfer factor that process of the present invention prepares

[0039] Properties: The main components of this product are low-molecular-weight active peptides and ribose. It is a white or light yellow solid in the frozen state. After thawing, it becomes a colorless or light yellow clear liquid, and there may be a small amount of peptide condensate precipitated.

[0040] pH value: Measured according to the appendix of the current Veterinary Pharmacopoeia of the People's Republic of China. At 20°C±2°C, the pH value should be 6.0-7.5.

[0041] Protein Qualitative: Take an appropriate amount of this product 2ml, add 20% sulfosalicylic acid reagent 2ml, shake well, the solution must not appear precipitation or cloudy reaction.

[0042] Sterility test: test according to the appendix of the current "People's Republic of China Veterinary Pharmacopoeia" 2005 edition, and it should grow aseptically.

[0043] Mycoplasma inspection: Accord...

Embodiment 3

[0052] The clinical application of embodiment 3 transfer factors

[0053] Experiment 1: Comparison of product content of different extraction methods

[0054] The transfer factor extracted by this patent method is compared with the main index of the transfer factor extracted by the comparative extraction method

[0055]

[0056] Experiment 2. The effect of transfer factor on broiler chickens

[0057] In this experiment, we not only used the comparative extraction method as a control, but also selected a mature transfer factor product currently available in the market as a control.

[0058] Select 0 day-old healthy broiler chickens, and then randomly divide them into 4 groups, 400 chickens in each group, and feed the basic diet (see appendix 1). Group 1 is a blank control group; Groups 2, 3, and 4 are test groups. - The immunization program of the group 4 is immunization with eye drop nasal drops at 1 day of age to immunize with Influenza 2886; at 7 days of age, attenuated N...

Abstract

The invention relates to the technical field of bioengineering, and particularly relates to a novel technology of a preparation process of a transfer factor. The preparation process comprises the following steps of: adding normal saline into a treated spleen of a laying hen, wherein the amount of the treated spleen of the laying hen is equal to that of the normal saline; homogenating by using a colloid mill and then adjusting a PH (Potential of Hydrogen) value to 3.5 at a temperature of 20 DEG C; repeatedly freezing and thawing for three times at a temperature of -20 to -25 DEG C; centrifuging and taking supernate for later use; taking residue and then grinding again, adjusting the PH value, repeatedly freezing and thawing for three times, centrifuging and taking supernate, and mixing the supernates; and carrying out virus inactivation and ultrafiltration, adjusting the PH value to 7.0, filtering and sterilizing, and the like to obtaining a transfer factor oral solution of the spleen of the laying hen. The transfer factor is mixture consisting of bioactive polypeptide and nucleotide, and has the functions of transferring a cellular immune function of donor cells to recipient cells, non-specifically enhancing ordinary cellular immunity, and improving an immunologic function of a receptor. The transfer factor is safe and non-toxic and is a novel immunomodulator. The transfer factor is mainly used for treating viral infections and autoimmune diseases. The immune effect can be obviously enhanced by using the transfer factor with a vaccine in a matched manner.

Description

technical field [0001] The invention relates to the technical field of bioengineering, in particular to a new technique for preparing transfer factor. Background technique: [0002] Transfer factor is a mixture of biologically active polypeptides and nucleotides, which can transfer the cellular immune function of the donor cell to the recipient cell, non-specifically enhance general cellular immunity, and improve the immune function of the recipient. Moreover, it is safe, non-toxic, and has no drug residue, and is a new type of immunomodulator. It is mainly used for the treatment of viral infections and autoimmune diseases. It can significantly increase the antibody titer and enhance the immune effect when used with vaccines. The general extraction process of transfer factor in the prior art is as follows: [0003] 1 Pretreatment of fresh tissue, after homogenization in a homogenizer, freeze and thaw repeatedly at -20°C-20°C for 3-8 times. [0004] 2 Centrifuge in a high-...
