Problems solved by technology

However, its practical application is restricted by the photobleaching properties of photosensitizers and short blood circulation life, and most of the photosensitizers are not ideal in water solubility, and they are easy to agglomerate or bind to proteins when the concentration is high, which ultimately affects the diagnosis and treatment effect.

Embodiment Construction

[0030] The technical solution of the present invention will be further described below in conjunction with specific embodiments and accompanying drawings.

[0031] The invention discloses a bionic recombinant lipoprotein / photosensitizer nano particle. On the basis of using biomimetic recombinant lipoprotein as the carrier, the selective targeting ligand RGD with better tumor homing and penetration ability is used to further improve the targeting of the carrier. RGD is a class of peptides containing a specific arginine-glycine-aspartic acid (Arg-Gly-Asp) sequence, which can specifically recognize integrin receptors on the surface of tumor cells or tumor neovascularization, and pass through the tumor site Protease cleavage produces CRGDK / R, which has high affinity with NRP-1 on the surface of tumor cells, and achieves deep tumor penetration through receptor-mediated endocytosis. Therefore, the nano-drug carrier coupled with RGD can deeply penetrate the tumor to reach the interi...

Abstract

The invention discloses a bionic recombined lipoprotein / photosensitizer nanoparticle, and a preparation method and an application of the bionic recombined lipoprotein / photosensitizer nanoparticle. Thenanoparticle comprises a phospholipid monomolecular layer, and a photosensitizer and cholesteryl ester that are enveloped in the phospholipid monomolecular layer, wherein RGD (arginine-glycine-aspartic acid) peptide modified apolipoprotein is embedded into the surface of the phospholipid monomolecular layer; and cholesterol is further distributed among phospholipid molecules of the phospholipid monomolecular layer. The nanoparticle is effectively accumulated at a tumor site through high permeability and a retention effect of a solid tumor and high affinity of the apolipoprotein and a scavenger receptor, and further achieves target deep penetration to the tumor using high affinity of RGD and an integrin receptor. The nanoparticle can generate active oxygen and high heat under irradiation of near infrared light, promotes quick release of the photosensitizer from lipoprotein, and achieves a synergic photodynamic and photo-thermal treatment effect; and at the same time, fluorescent lightgenerated by triggering of near-infrared wavelength light can effectively perform in-vivo diagnosis, so that the nanoparticle achieves double functions of target treatment and diagnosis.

Description

technical field [0001] The invention belongs to the technical field of pharmaceutical preparations, and in particular relates to a bionic recombinant lipoprotein / photosensitizer nanoparticle, a preparation method thereof, and an application in tumor diagnosis and treatment. Background technique [0002] Tumor has become a frequently-occurring disease that seriously threatens human life around the world. According to the latest research report, about 4.3 million people in China were diagnosed with cancer in 2016, of which 2.8 million died. Malignant tumors are regarded as a major disease that endangers human health because of their poor timeliness in diagnosis and treatment (theranostics), and have also become a hot spot in modern medical research. More and more researches are currently devoted to exploring diagnostic reagents and therapeutic drugs that target or regulate the tumor microenvironment. [0003] Lipoproteins are composed of lipids and proteins. The inner layer i...
