Embodiment 1

[0028] The extraction steps of embodiment 1, Scutellaria baicalensis polysaccharide are:

[0029] (1) Take the dried roots of the plant Scutellaria baicalensis and grind it through a 20-mesh sieve to obtain Scutellaria baicalensis powder;

[0030] (2) Take 10g of Scutellaria baicalensis powder, add 40mL of distilled water, and extract for 15min in a 60°C water bath with an ultrasonic power of 300W;

[0031] (3) Extract in hot water at 80°C for 1 hour, filter after cooling, add ethanol with a concentration of 95% to the concentrated solution to make the ethanol content in the concentrated solution reach 80%, and let stand at 4°C for 12 hours;

[0032] (4) Separate the precipitate and supernatant in a centrifuge at 1500r / min, collect the precipitate and dry it at 60°C to obtain 4.75g of Scutellaria baicalensis polysaccharide extract with a purity of 36.84%.

Embodiment 2

[0033] Example 2. In vitro porcine lymphocyte transformation experiment of Scutellaria baicalensis polysaccharide:

[0034] (1) Aseptically collect 20 mL of porcine anterior vena cava blood and put it into an anticoagulant tube containing heparin sodium.

[0035] (2) Dilute the anticoagulant blood and the diluent in the anticoagulant tube at a mass ratio of 1:1, and put the diluent and lymphocyte separation solution into an autoclaved centrifuge tube at a mass ratio of 1:2 to control the level The rotational speed in the centrifuge is 1850r / min, and the centrifugation is carried out for 30min.

[0036] (3) Absorb the lymphocyte layer of the white floc in the middle, put it in a clean high-pressure sterilized centrifuge tube, add cleaning solution and mix well, then centrifuge at 1500r / min for 10min; discard the liquid, add 1mL red blood cell lysate and blow evenly, Let it stand for 3 minutes, add cleaning solution to balance, and then centrifuge at 1500r / min for 10 minutes; d...

Embodiment 3

[0046] Example 3, Scutellaria baicalensis polysaccharide improves rat immunity detection experiment:

[0047]The experimental rats were divided into 5 groups, namely high-dose group (200mg / kg), middle-dose group (100mg / kg), low-dose group (50mg / kg), control group 2, blank control group, 6 rats in each group . Cyclophosphamide (70mg / kg) was administered to each rat in the high-dose group, the middle-dose group, the low-dose group and the control group respectively, and the blank control group was not administered, and the administration was continued for 3 days. After 3 days, the mice were given Blood samples were taken to measure immune indicators. Rats after blood collection were administered with Scutellaria baicalensis polysaccharide according to high, medium and low doses: high dose group 200mg / kg, medium dose group 100mg / kg, low dose group 50mg / kg, control group 2 and blank control group were not administered . 3d, 5d, and 7d after the administration, the rats were blo...

Abstract

An application of baikal skullcap polysaccharides in preparation of a veterinary drug is disclosed. The baikal skullcap polysaccharides are used for preparing an additive in a veterinary drug enhancing immune functions of animals, and are prepared through extraction of baikal skullcap plants. The concentration of the baikal skullcap polysaccharides in the veterinary drug is 31.25 [mu]g / mL to 4 mg / mL. Pig lymphocyte transformation tests show that the baikal skullcap polysaccharides can promote lymphocyte transformation, can increase contents of the total protein (TP), serum albumin (ALB), serumglobulin (GLOB), lymphocytes and monocytes in serum of rats with low immunity so as to restore body conditions of the rats, thus proving that the baikal skullcap polysaccharides have effect of significantly enhancing animal body immune functions and can be used in a veterinary drug enhancing animal body immune functions.

Description

technical field [0001] The invention belongs to the technical field of preparation of veterinary medicine in animal medicine, and specifically relates to the application of Scutellaria baicalensis polysaccharide in the preparation of veterinary medicine. Background technique [0002] Entering the 21st century, my country's animal husbandry industry is gradually intensified and scaled, and people's requirements for animal food safety are increasing. Chinese veterinary medicine is expected to become an important substance to ensure animal food safety and prevent animal diseases. Extracting active ingredients to prepare veterinary drugs has become a hot research topic in recent years. Modern molecular biology, physics and phytochemistry have achieved rapid development in recent decades, and the analysis and extraction techniques and methods of active ingredients in traditional Chinese veterinary medicine have made great progress. High performance liquid chromatography, mass spec...
