The method comprises the following steps: (1) pretreating raw materials; (2) carrying out reflux concentration; (3) preparing a sample: adding an obtained liquid extract into column chromatography silica gel and ethanol, stirring, and drying by distillation until a dispersed sample is obtained; and (4) carrying out column chromatography separation: putting the column chromatography silica gel into a chromatographic column, adding ethyl acetate to obtain a flowable solid-liquid mixture, adding the dispersed sample into the chromatographic column, eluting the chromatographic column with ethyl acetate, equivalently collecting all the fractions, then confirming whether lemon phenol is completely discharged ornot, enriching a part of fractions containing lemon phenol, and evaporating to obtain lemon phenol. The method for extracting lemon phenol from Desmodium caudatum is simple, easy to operate and few in step, and industrial production is facilitated. And the used solvents are few in variety and can be completely recycled. And the yield of lemon phenol in Desmodium caudatum is improved.
